Roger Federer was the two-time defending champion, but lost in the semifinals to Novak Djokovic.

Djokovic won in the final 4–6, 6–4, 6–3, 7–6^(7–2), against an unseeded Jo-Wilfried Tsonga (ranked no. 38) playing in his first ATP final. It was the first Grand Slam title of Djokovic's career, and the first of his eventual six Australian Open titles. It was also the first Grand Slam final since the 2005 Australian Open that did not feature either Federer or Rafael Nadal, who were beaten by Djokovic and Tsonga, respectively, in the semi-finals.^[1]^[2] This ended Federer's run of a record 10 straight Grand Slam finals. He would go on to make the next 8 Grand Slam finals, for a run of 18 Grand Slam finals in a span of 19 tournaments.
