County routes in Genesee County, New York, are not posted on route markers. However, the number is given, along with house numbers, often on intersection blade signs. There is no apparent numbering pattern. County routes generally comprise one or more named roads, or portions thereof, strung together to form a single continuous unit. These usually extend for some distance, connecting primary highways in two or more towns. The majority of the county's roads are assigned to the towns. Nowhere does any county route wholly enter either a village or the city of Batavia. Routes 41 through 52 are listed below.

County Route 41 is a short north–south route located in the town of Byron named South Holley Road. CR 41 starts at North Bergen Road (CR 14) and heads due north for just over a half-a-mile. There it reaches the Orleans County line where the CR 73 designation takes over. The roadway keeps its name north into Orleans County.

County Route 42 is an overall north–south route split into two segments by part of Bank Street Road (CR 13). It starts out running east–west as Walkers Corners Road at the intersection of CR 19 and CR 19A. From there, CR 42 heads west until it reaches Starowitz Road, onto which it turns north. It soon enters the town of Elba, where the southern segment of CR 42 ends at Bank Street Road (CR 13). The designation resumes 0.15 miles (0.24 km) northeast of there as Transit Road. As Transit Road, CR 42 heads due north for the remainder of its route, intersecting NY 262 (Byron–Elba Road) and ending at Watson Road, where county maintenance ends.

County Route 43 is a very short north–south route in the northwestern part of the town of Alabama. CR 43, named Salt Road, is only a quarter of a mile long and runs from NY 77 (named Lewiston Road and maintained by Genesee County as CR 12) north a short distance to the Orleans County line where Salt Road continues as CR 52.

County Route 45
County Route 45 is an east–west route in the towns of Alexander and Darien named Attica Road. It begins at NY 238 in the town of Darien and heads southeast on a parallel routing to NY 238. The route eventually crosses into the town of Alexander at an intersection with Alexander–Darien Townline Road (CR 36), and CR 45 comes to an end at another junction with NY 238 just north of the village of Attica. Most of CR 45 is separated from NY 238 by the Southern Tier Line.

County Route 46 is an east–west county route located in the town of Batavia, north of the city of Batavia. CR 46 begins at NY 98 (Oak Orchard Road) and heads east as West Saile Drive. At 0.71 miles, CR 46 crosses State Street Road and changes its name to East Saile Drive. As the roadway heads east, it passes along the southern edge of the Genesee County Airport for much of the rest of its length, before ending at Bank Street Road (CR 13).

County Route 47 is a north–south county route named Putnam Road. CR 47 begins at Creek Road (CR 1) north of the hamlet of East Alexander, and from there heads north-northeast to its end at Francis Road (CR 38) just over the Batavia town line. Putnam Road itself continues northeast as part of CR 38.

CR 48 starts Bank Street Road (CR 13) and heads west, passing south of Genesee Community College before reaching its end at Batavia–Stafford Townline Road (CR 19) on the Batavia–Stafford town line.

County Route 49 is a fairly short east–west route named Raymond Road in the town of Bethany. CR 49 starts at the intersection of Linden and Bethany Center roads (CR 15). It heads east from here, running along the northern border of the Genesee County Park and Forest. Memory Lane (CR 52) is the only road CR 49 intersects and allows access to the park. At 1.49 miles, CR 49 reaches its eastern end at East Road (CR 35).

County Route 50
County Route 50 is a north–south county route located in the towns of Pavilion and Le Roy named Perry Road. It begins just north of the Wyoming County line at an intersection with NY 63 and the northern terminus of NY 246 (Perry Road). From here, CR 50 heads due north through Pavilion, running parallel to the east of NY 19 for its whole length. At just over 3 miles, CR 50 intersects US 20 (Broadway), and continues heading north, into the town of Le Roy. At almost 6.4 miles CR 50 comes to its north end at NY 19 (Warsaw Road), southwest of the village of Le Roy.

County Route 51
County Route 51 is a north–south county route primarily located within the town of Darien and crosses into the town of Pembroke before ending. CR 51 is named Colby Road for its entire length and begins at a four-way intersection with US 20 (Broadway) and Tinkham Road. CR 51 heads due north from here intersecting only local and county roadways, and crossing into the town of Pembroke just before it reaches NY 33 (Genesee Street) where the county route has its north end.

County Route 52 is the highest numbered route in the county. It is an east–west route located within the town of Bethany named Memory Lane. CR 52 is a county route that runs through the Genesee County Park and Forest. It starts its west end at Bethany Center Road, just south of the Bethany Center Road and Raymond Road (CR 49) intersection. CR 52 heads east into the park, and turns south to run through the southern section and then turns back to the north to its exit at Raymond Road. In all, CR 52 is just under 2 miles in length.
