Route description

Franklin County

State Route 16 begins at the Alabama State Line, continuing as State Route 79 in Alabama. It continues north to Winchester, where its first major junction with U.S. Highway 64 and State Route 15 is located. In Winchester, it becomes concurrent with U.S. Highway 41A, which would last for the rest of its route. For most of this concurrency, however, it is unsigned.

Coffee County

Continuing its concurrency with U.S. Highway 41A, State Route 16 enters Bedford County via Tullahoma. In Tullahoma, it encounters State Route 55, which links Tullahoma with nearby Interstate 24.

Bedford County

In Shelbyville, the route serves as the primary east-west corridor. It intersects U.S. Highway 231, the city's other primary corridor.

Rutherford County

In Rutherford County, State Route 16 meets State Route 99 in Eagleville.

Williamson County

Williamson, County is the location of the route's northern terminus, at a junction with U.S. Highway 31A, U.S. Highway 41A, and State Route 11.
