The 45th Annual Miss Puerto Rico Universe competition was held in the fall of 1999 in Puerto Rico. Zoribel Fonalledas won the pageant and represented Puerto Rico at Miss Universe 2000 in Nicosia, Cyprus.

• Miss Guaynabo, Zoribel Fonalledas, represented Puerto Rico at Miss Universe 2000 in Nicosia, Cyprus where she failed to make the semi-finals, ending Puerto Rico's three-year streak of consecutive placements in Miss Universe, from 1997 through 1999. She is also the daughter of Jaime Fonalledas, President and CEO of Empresas Fonalledas Inc., which owns Plaza Las Américas, the largest shopping mall in the Caribbean and one of the top retail and entertainment venues in the world.
• Miss Naranjito, Barbara Serrano, was a famous model in the island. She later competed at Miss World Puerto Rico 2000 where she placed as 1st runner-up and was later crowned Miss World Puerto Rico 2001. She competed at Miss World 2001 where she didn't classified as a finalist.
• Miss Carolina, Karla Carrion, later competed at Miss Puerto Rico Universe 2002 where she finished as Top 5.
