Church is a Buffalo Metro Rail station located in the 300 block of Main Street (just north of Church Street) in the Free Fare Zone, which allows passengers free travel between Erie Canal Harbor Station and Fountain Plaza Station. Passengers continuing northbound past Fountain Plaza are required to have proof-of-payment.

Church Station is the closest to the Buffalo Metropolitan Transportation Center located two blocks east at Ellicott and North Division Streets.

From June 24, 2013 to October 14, 2013 and since March 30, 2014, due to the NFTA's "Returning Cars to Main Street" project, Metro Rail is single tracking. From June 24, 2013 to August 8, 2013 and from July 21, 2014 to October 10, 2014, the outbound track from Allen/Medical Campus Station to Church Station is used, as the inbound track is redone. From August 11, 2013 to October 14, 2013, from March 30, 2014 to July 17, 2014 and since October 14, 2014, the inbound track from Allen/Medical Campus Station to Church Station is used, as the outbound track is redone. As a result, Church Station serves as the southern terminus for passengers traveling from University Station and the northern terminus for passengers traveling to University Station, meaning passengers that wish to continue to and from Erie Canal Harbor Station or Seneca Station must change trains at Church.
