The team later released on September 14, 2010, but signed with the practice squad the next day. Five days later, he signed on to the active roster. Brock would be released again on October 12, then put on the practice squad two days later. He was signed to the active roster on the October 27. In 2010, Brock made his NFL debut against the Denver Broncos and that year only played in three games and recorded only four tackles.

He got his first interception in the first game of the season versus the Seattle Seahawks and his second versus the Dallas Cowboys with an 18 yard return.
2012 Season
He later signed a one-year deal on March 12, 2012. In 2012, Brock played a big role on special teams especially the kickoff, that year he played all 16 games. That year he played in the Super Bowl. Brock, recorded 16 tackles and a key forced fumble against the St. Louis Rams, a divisional rival, Brock, created the forced fumble that gave the 49ers the ball back and took advantage of the turnover to put them up in the lead 21-17.
2013 Season
In the 2013 offseason, Brock signed a $1.323 million tender with the 49ers keeping him there until the 2014 season.[4][5]
On October 6, 2013, Brock scored his first Touchdown against the Houston Texans on an interception off Quarterback Matt Schaub. Later in the game, Brock recorded his 2nd interception of the game, making it his first multi-interception game in his career. Brock was named the teams 3rd Cornerback, replacing the injured Nnamdi Asomugha, who was later released by the team.
Just days after the game against the Texans, Brock was named NFC Defensive Player of the Week.
On November 22, 2013, Brock agreed to a four year extension with the 49ers worth $16 million including $7 million in guaranteed money.[6]
When Tarell Brown went down with a rib contusion in week 11 in a controversial loss against the New Orleans Saints, Brock took over as duties as #2 cornerback. Since taking over these duties he hasn't looked back playing like a star corner in the end of the season heading into the playoffs.
The play that Brock will be forever remembered for happened in Week 16, on Monday Night Football against the Atlanta Falcons. It was a rematch of last years NFC Championship game, and it was the last regular season game at Candlestick Park. The (4-10) Falcons took the early lead and were up 10-3 at half time. The 49ers got up to a 27-17 lead with just over 5 minutes left in the 4th Quarter. Matt Ryan threw a 2 yard touchdown to Tony Gonzalez with 2:09 remaining to make it a 27-24 game. Atlanta recovered the onside kick and completed 2 quick passes to get down to the 49ers 10 yard line. Brock came in and broke up the pass which was picked off by NaVorro Bowman who returned it 89 yards to seal the game. Brock then finished the game with an interception in the end-zone as time expired.
