The Isthmian League 2009–10 was the 36th season of the Isthmian League Cup, the league cup competition of the Isthmian League. 66 clubs took part. The competition commenced on 20 October 2009 and finished on 24 March 2010.

The two clubs to have made it through the Semifinals reached the Final.
The final was held on Wednesday, 24 March 2010 at Imber Court, home of Metropolitan Police. The winners were Leatherhead, who beat Wealdstone by four to three on penalties after a nil–nil draw in front of six hundred and eighty-eight spectators.
