Now in its 50th year, Wayne's varsity football team has won several conference championships and has played in the statewide playoffs at least 10 times. In 1999, the team made it to the Division I state championship game. In 2005, the team was consistently ranked in the top 10 teams of Ohio.
Wayne's principal rival is Centerville High School. In 1981, Wayne's defeat of Centerville's varsity football team ended Centerville's six-year 53-game winning streak. "Fifty-three was all they'd see!" and "Fifty-four, there'd be no more!" were popular phrases chanted at Centerville games.
The Wayne men's track team won the Division I boys state team title in 1995. This was the first time in school history that an athletic team from Wayne had won a state title. The men won the state team title again in 2000.
- The mascot for Wayne athletics is the Warrior, depicted by an image of a Native American. When turned upside down, it forms the image of an eerily grinning clown. The logo is unique to the school, and is sometimes used as a symbol to represent the entire school district. The image of the warrior can be found painted on some walls in the school.
- Wayne's colors are red, white, and black.
- Wayne's fight song is the Washington and Lee Swing.
- Wayne is a member of the Greater Western Ohio Conference. Previously it was a member of the Western Ohio League.

Oh, when the Wayne High team all fall in line
We're gonna win this game another time
And for the dear old school we love so well
So for the Big Red team we'll yell, yell, and yell
And we will fight, fight, fight for every game
And set our standards on high for Dear Old Wayne
And we will roll those rivals on the side, on the side
Rah, Rah, RAH! YAY, WAYNE!
