U.S. Highway 72 (US-72), internally designated by the Alabama Department of Transportation as State Route 2 (SR-2), is a major east-west state highway across the northern part of the U.S. state of Alabama.

SR-2 and US-72 intersect with the following major junctions in Alabama:
- Natchez Trace Parkway west of Cherokee
- U.S. Highway 43, State Route 13, and State Route 17 at Muscle Shoals. These routes overlap until they split west of Killen, except for State Route 17 which splits at Florence.
- U.S. Highway 72 Alternate and State Route 20 at Muscle Shoals. State Route 20 overlaps until it splits at Florence.
- State Route 101 at Elgin
- U.S. Highway 31 and unsigned State Route 3 south of Athens
- Interstate 65 south of Athens
- State Route 255 at Huntsville
- State Route 53 at Huntsville
- U.S. Highway 231, U.S. Highway 431 and unsigned State Route 1 at Huntsville. These routes overlap from University Drive to Sparkman Drive.
