U.S. Route 340 in Maryland parallels the route of the old Jefferson Pike from Weverton to Frederick, meeting US 15 near the town of Jefferson.

The route is known as the Jefferson National Pike.

The route enters Maryland over the Potomac River, downstream from Harpers Ferry, West Virginia. After intersecting a local road the route immediately widens into a four-lane divided highway. It curves to the east and intersects the southern terminus of MD 67 1.6 miles from the river. Continuing east the route intersects the western terminus of MD 180 and upgrades into a high-speed four-lane freeway.

US 340 serves as a major east-west route through southern Frederick County, intersecting MD 17 north of Brunswick, and MD 180 for the second time near Slabtown. Now running due east the route serves as a bypass of Jefferson, before merging with US 15 a few miles east of the town.

Both routes now turn north and continue towards Frederick, intersecting Interstate 70 outside the city. US 340 (and the freeway) terminates at US 40 in downtown Frederick, joining with the parent highway via a partial cloverleaf interchange within the Frederick Triangle [2], while US 15 merges with US 40 and runs north along the Frederick Bypass. The physical roadway continues north as an at-grade city street, S. Jefferson St.

US 340 once followed Jefferson Pike, now part of MD 180, to Frederick, before being switched to its existing alignment in 1970. The route once ran north of its existing terminus via S. Jefferson St. to terminate at US 40 Patrick St., now part of MD 144.
