U.S. Route 222 Business is an auxiliary route of U.S. Route 222 located in Reading, Pennsylvania. Its northern terminus is at US 222 north of Laureldale. The southern terminus is at US 222 near Mohnton. US 222 Business is the only auxiliary route of U.S. Route 222 in Pennsylvania.

Business 222 starts at an interchange with US 222 north of Laureldale. Then Business 222 becomes Allentown Pike, a local road . Then becomes The "5th Street Highway" and has an interchange with Pennsylvania Route 12. It enters Reading and becomes 5th Street. 5th Street goes into downtown Reading with both northbound and southbound of Business 222. Then in Reading before entering the downtown area, it meets with Pennsylvania Route 61. In downtown Reading it meets with the westbound of Business U.S. Route 422 that is called Washington Street. After going through downtown, it has an intersection with the eastbound section of Business 422 that is Franklin Street. Then Business Route 222 becomes Lancaster Avenue and crooses the Schuylkill River. There is an interchange with U.S. Route 422 and then an intersection with Pennsylvania Route 10 just south of U.S. Route 422. About a mile from PA-10, Pennsylvania Route 625 meets with Business U.S. Route 222. It leaves Reading and enters Shillington. Later the route merges with Pennsylvania Route 724. Business 222 ends at an interchange with U.S. Route 222 near Mohnton.
