[edit] Sypnopsis
This time, Dr. Agaton will play as a pediatrician, who decided to seek greener pastures in the US by becoming a nurse. As it is his personal mission to help other people, Agaton gradually made his way up, and soon, he was recognized as one of the best nurses in the hospital he's working in! However, it seems that fame and fortune are unable to fill the longing in Agaton's heart. A pang of homesickness sets in, and he eventually decides to go back to the Philippines, and take a vacation with his Tita Ocs, his childhood guardian, while his own parents were in the States. Will he find what he's looking for here in the country?

- Aga Muhlach as Doc Agaton Abogado - a pediatrician. As a child, Aga had asthma. Add to that his father's frustration in becoming a doctor, so he was more than convinced to take the road to medicine. His personal mission is to help the people around him. However, he decided to seek greener pastures in the US by becoming a nurse. Soon, he becomes known as one of the best nurses in the hospitals he works in, but despite his fame and fortune, it seems as if he's still incomplete. One day, he sees a video of the Philippines, and gets homesick! A such, he decides to take a vacation, reconnects with his Aunt Octavella, who played as his guardian while he was growing up and his parents were still in the States, and heads straight to her community, Brgy. Tabuneknek.
- Nova Villa as Octavella Flores or Tita Ocs - Her husband has been mysteriously missing for 35 years already, so she became a Jill of all Trades so that she can provide for her child. However, her kid left the country for a foreigner, leaving Butching in Tita Ocs' care. This workaholic can't get to stop herself from working, and is the only sister of Aga's mother. She hates Major Ret to the bone, and is currently a pedicab driver. She also accepts plumbing and electirc fan repairs. She still has a soft spot for her husband, who apparently looks like Piolo Pascual.
- Roderick Paulate as Major Ret Retualo. This Forced Retired Major of the Philippine Army is still involved with classified missions,although no one believes it, as he tends to exaggerate his stories. This man had always dreamt to become a soldier, as he came from a family of one. This strict disciplinarian is very authoritative, to the point that he's feared by the community, which he takes pride in. He plays both mom and dad to his little sister Greta, and his son, Jake. He's overprotective as a father, supportive as a mother (since his son lost his mother at an early age) and a best friend as a brother.
- Bayani Agbayani as Buddy - the animal lover. His father is a conservationist, and when Buddy was five years old, he was brought to an expedition, and got abducted by an Orangutan! It turns out that Buddy reminded the gorilla of her son, who just died! As such, Buddy lived in the colony since then, making him rather close to animals. Always searching for his roots, Buddy tries to find love, but it seems that had found his match in Cita, an anthropologist's daughhter, whom he rescued from a bunch of monkeys! Cita gets smitten at this knight in shining armor, while Buddy reciprocates her love as she's the first woman he has laid his eyes on! They then move to the city, and Buddy eventually gets to own a pet store, beside Major Ret's sporting goods store. This animal rights advocate has a monkey with him in tow, lovingly named Kokong. In time, he feels that Cita isn't the right woman for him.
- Pokwang as Cita - whom Buddy rescued in the wilderness. The two made a vow to be with each other forever, which is something that she holds dear. She has blind love for Buddy, so she loves everything he loves, and hates everything and everyone, that he hates! She's rather tactless as well, and never seems to run out of words to say.
- Celine Lim as Butching - the "baby ng bayan". This curious seven year-old brings an infectious cheer in the community, and never hesitates to ask questions. She helps out with Tita Ocs in recycling at home, but sometimes conflicts with her Ate Eda due to their age gap. Meanwhile, Tito Aga seems somewhat like a stranger to her, since he left for the States when she was still a baby, but she looks up to him nonetheless.
- Eda Nolan as Eda - Kikay Girl . This 16 year-old got a college scholarship, so she went to Manila to reside with Ocs, as she is her mother's best friend. She knows the latest trends among teens, but she doesn't have them since she can't afford those. This fashionista is the official arranger of the plaza during fiestas, and has a thick Cebuana accent. She hides her being intelligent so people won't think she's a nerd. She respect Ocs a lot, although she's not fond of the old stuff Tita Ocs keeps. She has a crush on Aga, and sometimes clashes with Butching.
- Precious Lara Quigaman as Greta - Ret's younger sister/ Aga's girlfriend. A board passer, she's just waiting for her application to work as a nurse abroad, and works as Doc Aga's assistant for the meantime. Because she came from a line of soldiers, she enjoys physical activities, like sports and martial arts. She never fails to put sense in her Kuya's mind, and always defends him in front of other people. However, she resists her Kuya's overprotectiveness, since she knows she can take care of her self, and is Jake's "kabarkada slash ate".
- Jake Cuenca as Jake - Major Ret's son. Cheerful and accommodating, he loves to party, although he's quite disciplined as well. He defends his dad's strictness to the tenants, and has been used to the "military way" ever since he was a child. This music lover dreams of becoming a rock star. However, his father wants him to be in the military, and Jake doesn't want to disappoint him, which puts him in a predicament.
- Kokong - is Buddy and Cita's pet monkey, and Buddy's companion in the pet shop. This monkey seems to act like a human, and is sometimes left to guard the pet shop.
