Shire of Cook is a Local Government Area in Far North Queensland, Australia. The shire covers most of the eastern and southern parts of Cape York Peninsula, the most northerly land form on the Australian mainland.
Towns in the shire include Marton, Laura, Lakeland, Coen, Ayton, Rossville and Portland Roads as well as the administrative centre, Cooktown. There are also some inhabited islands in the shire such as Lizard Island.
The shire before 2007 contained a number of Aboriginal Council areas, such as Hope Vale, Lockhart River, Napranum and Mapoon. These are now separate councils.
The shire is home to two World Heritage Sites, the Daintree National Park and the Great Barrier Reef.
