Route 401 is a numbered State Highway running 2.2 miles in Rhode Island.

- Note: All RI 4xx highways were a southern terminus for RI 4 at various stages of its construction. All 4xx routes connect RI 4 to US 1.
Route 401 takes the following route through the State:
- East Greenwich: 2.2 miles; Route 2 to U.S. Route 1
- Division Street and First Avenue

Route 401, from just east of Cedar Avenue to First Avenue, runs on the Warwick/East Greenwich town line with Route 401 Eastbound running in East Greenwich and Route 401 Westbound running in Warwick.
