Phrynomedusa vanzolinii is a chordate, or more specifically an amphibian, in the Hylidae family. It is endemic to Brazil. Its natural habitats are subtropical or tropical moist lowland forests, subtropical or tropical moist montanes, and rivers. It is threatened by habitat loss.

Phrynomedusa vanzolinii is classified as data deficient, meaning more data is needed on the species for categorization, by the IUCN Red List of Threatened Species, the world's most comprehensive inventory of the global conservation status of plant and animal species maintained by the International Union for Conservation of Nature and Natural Resources. Phrynomedusa vanzolinii's population trend was unknown at last assessment.

This species is currently known only from Terespolis, in the state of Rio de Janeiro, and Boraceia, in the state of So Paulo, Brazil. Its distribution is not well known. It probably occurs more widely than current records suggest, especially in areas between the two known sites.
