Paolo Emilio Cardinal Rondinini was born in 1617 year to Alessandro Rondinini and Felice Zacchia. Grand-nephew on his mother's side of Cardinal Paolo Emilio Zacchia and grandson of Cardinal Laudivio Zacchia.

He studied in the University of Perugia and then became a cleric in the Apostolic Chamber in 1637 year.

Paolo Emilio Rondinini was created cardinal deacon in the consistory of July 13, 1643 by Pope Urban VIII and opted for the deaconry of S. Maria in Aquiro.He participated in the Papal conclave of 1644.

He was elected Bishop of Assisi on May 5, 1653. He participated in the Papal conclave of 1655. Opted for the deaconry of S. Giorgio in Velabro in 1655 year amd later for the deaconry of S. Maria in Cosmedin in 1656. Cardinal Rondinini participated in the Papal conclave of 1667. On April 30, 1668 he was promoted to the order of cardinal priests and opted for the title of Sant'Eusebio.
He died on September 16, 1668 in Rome.
