Er, how kan the Klenow Pol I fragment translate from a nick onwards? The Klenow fragment doesn't have 5' 3' exonuclease activity, hence it cannot remove the downstream nucleotides.

And a question concerning the second part: when DNase I nicks the dsDNA strand, it does so at random places, thus on both sense and antisense strands. But during polymerisation by Pol I, doesn't the progressive 5' 3' exonuclease eventually lead to a ds break?
