New York State Route 65 is a north-south state highway in the counties of Ontario, Livingston and Monroe in New York. The southern terminus of the route is at NY 5 and US 20 in West Bloomfield. The northern end of NY 65 is nearly 19 miles to the north at an intersection with NY 96 in Brighton.

NY 65 begins as Ontario Street in West Bloomfield. Heading north, NY 65 enters Livingston County for 1/4 of a mile before crossing into the village of Honeoye Falls in Monroe County. In the village center, NY 65 turns west onto East Street, crossing the Honeoye Creek (a tributary of the Genesee River), then returning on a northerly alignment as it turns onto North Main Street. NY 65 then becomes Clover Street outside of the village and retains this name until its terminus at NY 96 in Brighton 13 miles to the north.

Three miles to the north of Honeoye Falls, NY 65 intersects NY 251 in the Mendon hamlet of Rochester Junction, named for the Lehigh Valley Railroad spur that went to downtown Rochester and returned to the mainline at this point.
Continuing towards Rochester, NY 65 passes over the New York State Thruway (Interstate 90), intersects the eastern terminus of NY 253 and meets NY 252 (all in the town of Pittsford) as well as NY 31 in Brighton before reaching its northern terminus at NY 96.

NY 65 was assigned in the 1930 renumbering to a previously unnumbered roadway between NY 254 (now U.S. Route 20A) in Honeoye and NY 15 (modern NY 96) in Brighton. Between Honeoye and West Bloomfield, NY 65 was routed on what is now the northernmost portion of County Road 37.[2] NY 65 was truncated to its current southern terminus in West Bloomfield between 1948 and 1968.[3][4]
NY 65 used to be part of a concurrency with NY 253 in the town of Pittsford. Prior to the 1980s, NY 253 turned north onto NY 65 at its present eastern terminus and followed NY 65 north to Calkins Road, where NY 253 turned east and continued to the north towards Pittsford, East Rochester and Penfield.[5] In 1983, NY 253 was truncated to its present eastern terminus at NY 65 and the section of NY 253 from NY 96 north to NY 441 in East Rochester and Penfield was designated NY 153.[6]
