New York State Route 53 is a north-south state highway in Western New York. NY 53 is within the Finger Lakes District of New York throughout its existence between its northern terminus at NY 21 in the village of Naples in southern Ontario County and its southern terminus at NY 415 in the Bath hamlet of Kanona in northern Steuben County.

In the hamlet of Kanona, NY 53 begins at an intersection with NY 415, the former routing of NY 15 through the Southern Tier. NY 53 occupies Main Street for a short distance before turning north onto an unnamed road. Just northeast of Kanona, NY 53 interchanges with Interstate 86, concurrent with NY 15 and NY 17 here, at exit 37. This interchange is located less than a mile from the southern terminus of Interstate 390 at exit 36.
North of I-86, NY 53 maintains a northeast routing through the town of Wheeler. Near Stickneys, the route begins to turn due north as it enters Prattsburgh. NY 53 takes on a northeast alignment upon exiting.
Roughly three miles outside of Prattsburg, NY 53 turns to the east, paralleling the county line for four miles before turning to the north and crossing into Ontario County near Ingleside.

NY 53 runs for only two miles in Ontario County, maintaining a north-south alignment between the county line and its northern terminus at NY 21.

NY 53 was assigned in the 1930 renumbering to its current alignment from Kanona north to Prattsburgh. Past Prattsburgh, NY 53 continued north on Italy Hill Road, Italy Friend Road, and modern NY 247 before terminating at U.S. Route 20/NY 5 east of Canandaigua. What is now NY 53 from Prattsburgh to Naples was designated NY 247.[2] The routings of NY 53 and NY 247 were flipped by 1948; however, NY 247 was truncated to end at NY 364 in Potter.[3]
