New York State Route 289 is a short state highway in northern New York, USA. This highway is a north-south road that connects the south part of the Town of Henderson to the central part of the adjacent Town of Ellisburg and is entirely within Jefferson County.

The northern terminus of NY 289 is at New York State Route 178 between Roberts Corner and Thomas Settlement in the Town of Henderson. The southern terminus is at New York State Route 193 in the Village of Ellisburg near the center of the Town of Ellisburg.

NY 289 begins in the village of Ellisburg, New York at NY 193. It travels north along Main Street. As it leaves the village of Ellisburg, NY 289 travels through a rural area and open fields.

NY 289 then approaches the hamlet of Belleville. In Belleville, NY 289 intersects with Jefferson CR 75. At this intersection heading north, Jefferson CR 75 is to the east and continues north. NY 289 continues to the northeast, following the Sandy Creek.

NY 289 passes the back road to the Belleville-Henderson Central School as it heads northeast towards its northern terminus at NY 178, about 3 miles west of Interstate 81's interchange 41 (At NY 178).
