Maryland Route 404 is a major highway in the Eastern Shore region of Maryland. It runs from Wye Mills to the Delaware state line where it continues to Five Points, near Rehoboth Beach as Delaware Route 404. The two state highways together cross the width of the Delmarva Peninsula and serve to connect the cities west of the Chesapeake Bay (via US 50) with the beaches in Delaware.

Most of Maryland Route 404 is a two-lane road. The only four-lane, divided section of Maryland Route 404 is the bypass of Denton. The former alignment of Maryland Route 404 in Denton is signed as Maryland Route 404 Business. Another former segment of Maryland Route 404 through the towns of Queen Anne and Hillsboro is known as Maryland Route 404 Alternate.

Historically, the route's western terminus was at Maryland Route 2 in Anne Arundel County. It crossed the Chesapeake Bay via the Sandy Point-Matapeake ferry, roughly where the Chesapeake Bay Bridge is now and ran along present day Maryland Route 8, Maryland Route 18, and Maryland Route 662.[2] The Chesapeake Bay Bridge was originally to be designated as part of MD 404, but in 1948 US 50 was extended onto the Eastern Shore, replacing 404 and cutting it back to its current terminus in Wye Mills.

As of 2007, the Maryland State Highway Administration is working on improvements to the highway in order to provide relief to travelers driving to the ocean resorts. These improvements include the widening of the route and an interchange with US 50 as part of a separate planned project to convert the intersections along US 50 between the split with U.S. Route 301 and MD 404 to interchanges.
