Maryland Route 331 is a two-lane rural road passing through three Mid-Shore counties on Maryland's Eastern Shore.

Traveling a total of 30 miles starting at the intersection with U.S. Route 50 in Easton, it crosses over the Dover Bridge over the Choptank River, through Bethlehem, Preston, and Hurlock, before ending at Vienna on the Nanticoke River.

Route 331 passes by the original location of colonial settlement in Caroline County as well as Linchester Mill. The mill provided grain to Continental Army troops in the Revolutionary War and has operated since 1682.

Before 1940, all of MD 331 was part of U.S. Route 213. After the opening of the Choptank River Bridge in 1940, US 213 was diverted to cross the Choptank River via the bridge; after 1949, US 50 replaced US 213 across the bridge.

Between Rhodesdale and Vienna the route follows an abandoned rail line.
