The sixth season of MADtv aired from October 7, 2000, to June 23, 2001, with 30 episodes.

MADtv began its sixth season with several cast changes. Pat Kilbane and Phil LaMarr left the show at the end of the fifth season. Returning repertory players Alex Borstein, Mo Collins, Michael McDonald, Will Sasso, Aries Spears, Nicole Sullivan, and Debra Wilson were joined by Nelson Ascencio (a featured player from last season) and newcomer Christian Duguay. Andrew Daly, Dannah Feinglass, Jeff Richards, and Stephnie Weir were added as featured players.

Cast members reprised many of their well-known characters and schticks. Alex Borstein and Will Sasso interviewed celebrities at red-carpet events, while Sasso performed celebrity impersonations of Robert De Niro and Fred Durst of Limp Bizkit. Mo Collins appeared as Doreen Larkin, Trina Moss, Lorraine Swanson, and Liz Whitman-Goldfarb. Debra Wilson and Aries Spears appeared frequently as Whitney Houston and Bobby Brown.

The cast also added several new characters. Michael McDonald portrayed the Depressed Persian Tow Truck Man (later named Mofaz the next season), an irritable American immigrant who uses pop culture references to illustrate his family's shortcomings. Stephnie Weir played Dot Goddard, a seven-year-old girl whose intelligence pales next to her twin sister, Karen (Alex Borstein), a child genius who has written several books, found cures for diseases and planned NASA expeditions. Michael McDonald and Mo Collins played Dot's parents, who treasure Karen but despise Dot. Stephnie Weir also unveiled the characters Mrs. Campbell, a curious old lady, and Dr. Kylie, an immature, party-loving doctor. Season five guests Seth Green and Susan Sarandon returned periodically during season six.

Christian Duguay impersonated Jay Leno, President George W. Bush, and *NSYNC member Joey Fatone. Duguay also played hyperactive preteen Edward and Loopgarue of "Loopgarue & Hulahoop," with Mo Collins as Hulahoop.

Jeff Richards soon left MADtv but later appeared on Saturday Night Live, where he became well-known for his "Drunk Girl" character during his three-year tenure. Nelson Ascencio and Dannah Feinglass also left MADtv at the end of season six.

The MADtv logo appears and the theme song, which is performed by the hip-hop group Heavy D & the Boyz, begins. Each member of the repertory cast, followed by the featured cast, is introduced alphabetically. The screen is divided into three live-action clips of each cast member performing recurring characters during the introductions. When the last featured cast member is introduced, the music stops and the title sequence ends with the phrase "You are now watching MADtv."

Refusing to accept losing the Presidential election, Al Gore (Michael McDonald) handcuffs himself to the desk in the Oval Office. Pro golfer Tiger Woods (Aries Spears) promotes his new restaurant for blacks, whites, and Asians. A flamboyant postal worker (Nelson Ascencio) annoys his customers.

On Dateline, Jane Pauley (Alex Borstein) interviews a high school principal (Michael McDonald) with immoral and illegal cures for teenage ills. A woman (Mo Collins) regrets online dating when she meets her date (Michael McDonald).

An episode of Gilligan's Island in Spanish shows Gilligan (Michael McDonald) and the gang preparing to leave the island where they're stranded. Michael McCleod (Will Sasso) and Jasmine Wayne-Wayne (Alex Borstein) recap their singing career and sing their newest hit, "This Is The Land That I Love". Loopgarue (Christian Duguay) and Hulahoop (Mo Collins) sings songs at a cineplex. Two crackheads (Mo Collins, Aries Spears) try to repair their house and their relationship. News reporters, sports announcers, and political figures predict the winner of Super Bowl XXXV before the game ends. A man (Will Sasso) who doesn't understand that his girlfriend (Alex Borstein) wants to break up with him resorts to stalking her. A parody of What Women Want focuses on the female protagonist (Nicole Sullivan) reading men's perverted thoughts. A lost clip from Men Are From Mars, Women Are From Venus features a discussion on whether Cybill Shepherd (Nicole Sullivan) is attractive to men. Darlene McBride (Nicole Sullivan) tries rapping in her new music video, a parody of Eminem's Stan featuring Dido (Stephnie Weir) as an unwed mother.

Jay Leno (Christian Duguay) interviews Jennifer Lopez (Debra Wilson) on The Tonight Show with Jay Leno. Edward (Christian Duguay) struggles hard working at a fast food joint. Julie Andrews (Mo Collins) shows a lost scene from Mary Poppins where she uses illegal Mexican immigrants to clean her young charges' room. Several guests think that the plumber is gay. Trina Moss (Mo Collins) thinks she may finally get laid. A couple (Alex Borstein, Michael McDonald) has a tough time discussing their problems.
#622 April 28, 2001 Tom Green, Green Day (musical guests) Unable to get one from Belma (Aries Spears) and Tovah (Debra Wilson), Bill Clinton (Will Sasso) gives himself a Reality Check. Tom Green preferrs to open the show by sucking a cow's udder rather than promote his new movie, Freddy Got Fingered. Ms. Swan (Alex Borstein) has an admirer (Andrew Daly). A driving instructor (Nicole Sullivan) with a criminal record gives a student (Michael McDonald) no choice but to obey her commands. Child prodigy Karen Goddard (Alex Borstein) shows off her new cloning machine while her dim-witted sister Dot (Stephnie Weir) babbles nonsense. Bring It On and Sugar & Spice combine in a parody. Will Sasso interviews wrestler-turned-actor The Rock at a press junket for the film, The Mummy Returns. Bob Newhart plays a psychotherapist with a bizarre cure for depression for a patient (Mo Collins).
