The twelfth season of MADtv aired from September 16, 2006, to May 19, 2007, with 22 episodes.

A few minor cast changes were made during season twelve. The only cast members who did not return were Stephnie Weir (who left midway through season eleven), Frank Caliendo (who chose not to renew his contract), and Daniele Gaither (who was let go by FOX executives). Frank Caeti and Nicole Randall Johnson, featured performers from season eleven, were promoted to repertory status. Lisa Donovan made her first appearance as a featured performer in the season's fourteenth episode.

Season twelve featured some format changes. Previous seasons featured a sketch before the opening credits, but this was dropped for season twelve episodes; instead, previews of sketches from that particular episode appear. As in the show's early years, sketches were shorter and were sometimes split into separate parts to prevent them from dragging (such as a three-part filler piece about apropos TV shows saved on the TiVos of Tom Cruise, Tommy Lee, and George W. Bush).

After Frank Caliendo's departure, political sketches about George W. Bush stopped appearing as often. Season twelve also featured more TV and movie parodies, celebrity-based sketches, one-off situational comedy sketches, with recurring character sketches comprising the rest. Still, MADtv continued to air vaguely political sketches such as the "Steve Jobs Presents the iRack" sketch, featuring Michael McDonald as the Apple executive unveiling the iRack at a trade show. Test audiences tell him that the "iRack is falling" and to "take things out of the iRack" (thereby riffing on the word as a pun for Iraq).

Michael McDonald reprised a character from season eleven, Larry Felmer, an obsessive fan who gets extremely angry when celebrities are mocked. Other returning characters were Crista Flanagan's Luanne Lockheart and Wendy Walker (Three Minute Meals), Nicole Parker's Disney Girl, Nicole Randall Johnson's Ka-Son, Keegan-Michael Key's Coach Hines, and Michael McDonald's Stuart Larkin.

New sketches in season twelve included a Korean soap opera spoof called Attitudes and Feelings Both Desirable and Sometimes Secretive, starring Bobby Lee. Attitudes and Feelings, spoken in fluent yet random Korean words and subtitled in comical broken English (the dialogue and subtitles are not related), consists of rather bizarre soap opera clichés about the twisted life of Dr. Poon Ji Sum, his sexual antics, and his enemy, Korean crimelord "Mr. President." Each sketch ended with a cliffhanger. (The sketches were usually shown about five episodes apart.) Lee also played Johnny (Xian Hi) Gan, a Korean millionaire who hosts a rather odd talk show where him and his assistant, Pongo (who seems to be in love with him) interview celebrities.

Animated sketches returned to MADtv after being absent for several years. Claymation also appeared for the first time in seven years with the recurring sketch "Celebrity Pets", in which pets of celebrities talk about their daily lives with their celebrity owners. The show also featured some non-sequitur claymation sketches such as "Red Snow" (a Happy Feet-meets-Saving Private Ryan movie trailer), a Valentine's Day cartoon where Cupid murders two would-be lovers with hollow point arrows, "Real Police Video: Immigration", "Cartoon Network Rejects", and a Christmas short where George W. Bush blackmails Santa into killing his enemies. MADtv also developed a popular recurring animation sketch called "Weekly News with Toby", in which first grader Toby (voiced by Frank Caeti) talks about his view on current events.

The twelfth season features a new title sequence with shots of the performers as they prepare for the show. The screen divides into three different live-action shots. When the theme music starts, the announcer introduces each cast member alphabetically. After the last cast member is introduced, the whole cast is shown in a live-action shot. The music stops and the title sequence ends with the phrase "You are now watching MADtv." The sequence also features a new announcer.

Lauren Conrad (Nicole Parker) goes through her usual dramatic histrionics in a parody of The Hills. Hundreds of immigrants pop out of a driver's trunk as he's being interrogated by the police on an episode of Real Police Videos (claymation short). Maury Povich (Michael McDonald) and Connie Chung (Bobby Lee) star in their own reality show. Ellen DeGeneres (Nicole Parker) becomes the new spokeswoman for adopting Iraqi children. BET premieres a music video in which Jordan Peele sings about his love for video games. The Dixie Chicks' lead singer Natalie Maines (Crista Flanagan) gets her comeuppance from her sisters (Nicole Parker, Arden Myrin) in a music video parody of "Not Ready To Make Nice". Tyler Perry (Keegan-Michael Key) reprises his role as Madea in a comedy film called Ladies' Night, co-starring other black male comedians such as Eddie Murphy (Nicole Randall Johnson) and Martin Lawrence (Jordan Peele) in drag. Stop-motion animated commercial for a terrorist Barbie doll. In a parody of Project Runway, designers must create the latest fashions for leprechaun models (three-part sketch). The hosts of Inside Looking Out (Nicole Parker and Jordan Peele) reminisce about the their first meeting at a roller rink in the 1980s. A yoga teacher (Bobby Lee) is unable to film his routine for a DVD because of an annoying pedestrian (Ike Barinholtz) and his dog at the beach. Billy (Bobby Lee) learns about the dangers of pedophiles on Internet chat rooms in a mixed parody of Dateline: To Catch A Predator and Sesame Street. Former presidential nominee John Kerry (Michael McDonald) tries to clear the air about his recently controversial "botched joke" (two-part sketch). Nelly Furtado gets hounded by Eugene (Keegan-Michael Key), who tricks her into making a bootleg for him. The Weekly News with Toby features Rush Limbaugh's recent comments about Michael J. #1205 November 11, 2006 Seth McFarlane, Eva Longoria In a parody of the A&E reality show Intervention, the MADtv cast confronts Bobby Lee about his exhibitionism. The Cartoon Network presents rejected cartoon pilots, including ones about a Korean superhero and an amputee superhero team. A manager (Michael McDonald) must deal with an insane prospective employee (Nicole Randall Johnson) who constantly plays the race card to justify her actions. Nelly Furtado (Nicole Parker) and Timbaland (Jordan Peele) star in a music video parody of "Promiscuous" about catching venereal disease. While watching a kung fu film, Bobby Lee performs various martial arts moves for a skeptical Keegan-Michael Key. In a Terry Gilliam-esque piece, Madonna's adopted child gloats how his life changed after moving to America. A trailer for Dreamgirls shows the movie copying other films and VH-1 documentaries about musicians and their struggles with fame. A lovestruck woman (Arden Myrin) visits the location where the Los Angeles Kings practice and hits on Sean Avery. Jovan Muskatelle (Keegan-Michael Key) frantically reviews the most controversial events of 2006, much to the chagrin of a local news reporter (Ike Barinholtz). Employees question if gorillas can learn martial arts to get the upper hand over the elephants, but the employee (Crista Flanagan) giving the speech was supposed to come up with a new brand of cola.
#1211 January 20, 2007 Carmen Electra Steve Jobs's (Michael McDonald) new iPhone gives him magic powers, much to the shock of the studio audience and Nicole Parker. On Sesame Street, Big Bird suffers from radiation poisoning after being exposed to a nuclear tank made by Oscar the Grouch. Tiger Woods (Jordan Peele) showboats while competing in a golf tournament. Two women (Crista Flanagan, Arden Myrin) can't stop laughing at an office joke about being "coffee twins." Check out what TV shows are on the TiVo playlists of famous people such as Tom Cruise, Tommy Lee, and George W. Bush (three-part filler piece). Carmen Electra is a special guest on The Korean Movie Review Show. The host (Frank Caeti) of a Discovery Kids show praises the Internet, then reveals how Dateline NBC arrested him for soliciting sex from a minor after responding to a sexually suggestive MySpace message. #1212 February 3, 2007 Shawne Merriman, Mary Lynn Rajskub After Ike Barinholtz and Bobby Lee aren't invited to the Super Bowl, they decide to play backyard football with San Diego Chargers defensive end/linebacker Shawne Merriman. In 1314, a Scottish warlord (Jordan Peele) can't convince his men that he was raped by the British. Despite skepticism from one of their friends (Frank Caeti), the Superstitious Knights (Keegan-Michael Key, Jordan Peele) perform victory dances to help the Chicago Bears win the Super Bowl. A news station reports on supposed terrorist attacks and AIDS-infected puppies. Mofaz the Depressed Persian Tow Truck Man (Michael McDonald) and Condoleezza Rice (Nicole Randall Johnson) grouse about how security-obsessed America has become. Ike Barinholtz, Keegan-Michael Key, and Michael McDonald discuss who will win this year's Super Bowl. Paula Abdul (Nicole Parker) reports live via satellite to dispel rumors of substance abuse, but she constantly confuses Keegan-Michael Key with the names of other actors. Unaired openings of Law & Order feature people discovering dead bodies during awkward situations and a garbage man (Jordan Peele) freaking out over a discarded lamp. A new store features piñatas shaped like minority suspects (courtesy of the L.A.P.D.) that kids can whack repeatedly. The presence of presidential nominee Barack Obama (Keegan-Michael Key) overshadows a speech by Hillary Clinton (Nicole Parker). While a man (Michael McDonald) proposes to his girlfriend (Nicole Parker) at an aviary, they are splattered with bird droppings. The Blind Kung-Fu Master (Bobby Lee) is sent to space to stop an alien princess (Crista Flanagan) from dominating the universe. Rapper Flavor Flav (Jordan Peele) recounts skewed versions of famous moments in African-American history, to the chagrin of Sidney Poitier (Keegan-Michael Key) (three-part sketch). #1215 February 24, 2007 Danny Bonaduce Keegan-Michael Key and Arden Myrin get robbed by gang members while showing off their Academy Awards apparel. Oscar-nominated actor Forest Whitaker (Jordan Peele) raps about his many film roles and how he deserves an Oscar, with additional flavor from rapper Snoop Dogg (Keegan-Michael Key).
