Louisiana Highway 621 (LA 621) is a state highway in Louisiana that serves Ascension Parish. It spans 6.87 miles in a west to east direction.

From the west, LA 621 begins at a junction with LA 73 just north of its diamond interchange with I-10 (Exit 173) near Prairieville. It heads east and intersects with US 61 (Airline Highway) just north of Gonzales. Continuing eastward, it intersects LA 44 before ending at an intersection with LA 431.
LA 621 is an undivided, two lane highway for its entire length.
