Route 321 began on 8 November 1997 as a replacement for the withdrawn eastern section of route 21, running between New Cross Gate and Foots Cray using single decker low floor Dennis Dart SLF / Plaxton Pointers from London Central's New Cross and Bexleyheath Garages.
In February 2002 the Bexleyheath allocation was withdrawn, with the whole route being run from New Cross Garage. During January 2003, the route was converted to double deck Volvo B7TL / Plaxton Presidents.
