- Jack Nicholson played the baby's father in "Opie Finds a Baby" and played the defendant in "Aunt Bee the Juror".
- George Kennedy was a police officer in "The Big House". Among his movie credits are Cool Hand Luke and Airport '77.
- Lee Van Cleef played Skip, a carnival worker and thief in episode 32, "Banjo Playing Deputy". Van Cleef was the villain Angel Eyes in The Good, the Bad and the Ugly.
- Jean Hagen played Elizabeth Crowley in "Andy and the Woman Speeder." She played the ditzy Lina Lamont in Singin' in the Rain. She also played Marjorie Williams on The Danny Thomas Show.
- Jackie Coogan played George Stevens in "Barney on the Rebound." A famous child film actor, starring with Charlie Chaplin in The Kid, Coogan was also Uncle Fester on The Addams Family.
- Everett Sloane appeared as Jubal Foster, the farmer/moonshiner in "The Keeper of the Flame". He also composed lyrics for the show's theme song; Griffith sang them on the show's soundtrack album, but the lyrics were never used on the show. Sloane is known for his role as Mr. Bernstein in Citizen Kane.
- Bo Hopkins played George in "Goober the Executive."
- Pat Hingle played Fred Gibson in "Wyatt Earp Rides Again." His movie appearances include the role of Commissioner James Gordon in Batman, Batman Returns, Batman Forever, and Batman & Robin.
- Sterling Holloway played Burt Miller, the traveling merchant in "The Merchant of Mayberry". A veteran of numerous films, Holloway was also the voice of Winnie the Pooh.

[edit] Gomer Pyle, U.S.M.C.
- Allan Melvin played different characters in eight episodes of The Andy Griffith Show. On the Andy Griffith Show spin-off, Gomer Pyle, he was Sgt. Carter's rival, Sgt. Hacker, the mess cook. Melvin is also noted for his roles as Sam Franklin on The Brady Bunch and Barney Hefner on All in the Family. In addition, Melvin directed some episodes of Andy Griffith, including Don Knotts' return appearance in 1966, "The Return of Barney Fife".
- Larry Hovis played Gilly Walker in "Goober Takes a Car Apart" and "The Case of the Punch in the Nose." Hovis played Pvt. Larry Gotschalk on Gomer Pyle as well as Sgt. Andrew Carter on Hogan's Heroes.

- Frank Cady played a drunk inmate named Luke in "The Rehabilitation of Otis" and Farley Upchurch in "The Legend of Barney Fife." Cady is best known for playing Sam Drucker on Green Acres. He also played the town drunk in the episode of Make Room for Daddy which was the pilot episode of The Andy Griffith Show.
- Hank Patterson played a hobo in "If I Had a Quarter Million Dollars." Patterson is best known for playing farmer Fred Ziffel.
- Mary Grace Canfield played Mary Grace Gossage in "A Date for Gomer." She portrayed Ralph Monroe.
- Alvy Moore played a kitchenware salesman in "A Baby in the House." Moore was Hank Kimball.
- Sid Melton played Pat Michaels in "The Hollywood Party." Melton was "Uncle Charley" Halper on The Danny Thomas Show and Alf Monroe on Green Acres.

- Barbara Eden played Ellen Brown, the title character of "The Manicurist". Eden starred in I Dream of Jeannie.
- Hayden Rorke played A.J. Considine in "Taylors in Hollywood." Rorke is best known for his portrayal of Col. Alfred E. Bellows, M.D.

- Richard Bull played Bill Lindsay in "Goober Makes History" and Mr. Jackson in "Opie's Piano Lesson." Bull is best known for playing Nelson "Nels" Oleson.
- Dabbs Greer played Councilman Dobbs in "Sheriff Barney." Greer also portrayed Reverend Robert Alden.
- Karl Swenson played Mr. McBeevee in "Mr. McBeevee." Swenson had the role of Lars Hanson.

- Jamie Farr played a gypsy named Gracos in "The Gypsies". Farr's most famous role is Max Klinger.
- William Christopher played Dr. Peterson in "A New Doctor in Town" and an IRS agent Mr. Heathcoat in the episode "Aunt Bee on TV" Christopher is best known for his role as Father Mulcahy. He also played Private Lester Hummel on Gomer Pyle: U.S.M.C.

- Edgar Buchanan played Henry Wheeler in "Aunt Bee's Brief Encounter". Buchanan is best known for playing Joseph "Uncle Joe" Carson.
- Charles Lane played Mr. Frisby in "Aunt Bee the Crusader." Lane portrayed J. Homer Bedloe.

- Bob Denver appeared in "Divorce, Mountain Style", as Dud Wash, the husband of Charlene Darling, who is jealous of her attraction to Andy.
- Alan Hale Jr. played "Big Jeff" Pruitt, the farmer who falls in love with Thelma Lou, much to Barney's dismay, in "The Farmer Takes a Wife".

- Rob Reiner played a printer's assistant named Joe in "Goober's Contest." Reiner is famous for playing Michael Stivic in All in the Family, and is the son of actor Carl Reiner.
- Howard Hesseman played a counterboy in "Goober Goes to an Auto Show" and Harry in "Sam for Town Council." Hesseman's best known roles were Dr. Johnny Fever on WKRP in Cincinnati and Charles P. Moore on Head of the Class.
- James Best played guitarist Jim Lindsey in "The Guitar Player" and "The Guitar Player Returns". He played Sheriff Rosco P. Coltrane on The Dukes of Hazzard.
- Buddy Ebsen played Dave Browne, the drifter hobo in "Opie's Hobo Friend". Ebsen is best remembered for his role as Jed Clampett on The Beverly Hillbillies.
- Jerry Van Dyke played bumbling musician Jerry Miller in "Banjo-Playing Deputy". He was considered as a permanent replacement for Don Knotts, who left at the end of season 5. However, Van Dyke opted to take a starring role in the flop TV series My Mother The Car instead. Years later, Van Dyke said he wished he had accepted the offer. He later portrayed Luther Van Dam on Coach.
- Bill Bixby played Ronald Bailey in "Bailey's Bad Boy". Bixby later starred in five TV series. During the course of his long career, he starred as Tim O'Hara in My Favorite Martian (1963–1966) on CBS, Tom Corbett, in The Courtship of Eddie's Father (1969–1972) on ABC, and Dr. David Banner in The Incredible Hulk (1978-1982) on CBS. He also starred in The Magician (1973) and Goodnight Beantown (1984). Bixby and Don Knotts would work together again in the 1975 family western The Apple Dumpling Gang.
- Gavin MacLeod played Sheriff Taylor in the Hollywood version of "Sheriff Without a Gun" in "Taylors in Hollywood" and a bank robber in an earlier episode "Crime Free Mayberry". Gavin also starred as Capt. Merrill Stubing in the The Love Boat and as news writer Murray Slaughter on The Mary Tyler Moore Show.
- Jack Albertson played Bradford J. Taylor in "Aunt Bee's Cousin." Albertson is best known for playing Ed Brown on Chico and the Man. His movie roles include Grandpa Joe in Willy Wonka and the Chocolate Factory, and Amos Slade in The Fox and the Hound.
- Jack Bannon played the announcer in "The Mayberry Chef." Bannon portrayed Art Donovan on Lou Grant.
- Ann Morgan Guilbert played Ella in "Aunt Bee's Cousin. Guilbert is best known for playing Millie Helper on The Dick Van Dyke Show and Yetta on The Nanny.
- Bing Russell played Mr. Burton in "The Loaded Goat." Russell was Deputy Clem Foster on Bonanza. Russell is also the father of actor Kurt Russell.
- Whitney Blake played Lee Drake in "Andy's Trip to Raleigh." Blake is best known for playing Dorothy Baxter on Hazel and for co-creating One Day at a Time with her husband, Allan Manings. Blake is also the mother of actress Meredith Baxter.
- Norman Alden played Tom Strongbow in "The Battle of Mayberry." Alden is best known for Aquaman in Super Friends and The All-New Super Friends Hour.
- Burt Mustin played Jud Fletcher in "Andy Saves Barney's Morale", "A Plaque for Mayberry", and "The Clubmen", and Sam Benson in "Opie's Newspaper." He was Gus the fireman on Leave it to Beaver.
- Leon Ames played Mr. Hampton in "The Senior Play." Ames played the role of Col. Gordon Kirkwood on Mister Ed.
- Roger Perry played George Jones in "Don't Miss a Good Bet." Perry is best known for playing Mr. Parker on The Facts of Life.
Morgan Brittany played Mary Alice, Opie's first date. She played on Dallas as Pam's sister, Katherine Wentworth.

- Don Rickles played traveling salesman Newton Monroe in "The Luck of Newton Monroe".
- Laurie Main played assistant publisher Robling Flask in "Helen, the Authoress." Main is best known for hosting and narrating for Welcome to Pooh Corner, as well as narrating for many "Disney Read-Along" records and cassettes.
- Leo Gordon played an ex-convict in "High Noon in Mayberry".
- Doodles Weaver played George Bricker in "Aunt Bee's Brief Encounter." Weaver is the brother of former NBC president Pat Weaver and the uncle of actress Sigourney Weaver.
- Jesse Pearson played singer Keevy Hazelton in "A Singer in Town." Pearson is best known for playing Conrad Birdie in Bye Bye Birdie.
- Joyce Van Patten played Laura Hollander in "Opie Steps Up in Class." Van Patten is the sister of actor Dick Van Patten.
- Guitarist Clarence White played a member of The Country Boys.
- Guitarist Herb Ellis played Bobby Fleet in two episodes from the first season.
