He became priest on the 17th of December 1960 with the order of Friars Preachers. In October 20 1983, Leonard Legaspi O.P. was appointed Archbishop of Nueva Caceres. He was also the first Filipino rector of the University of Santo Tomas Central Seminary, the first Vicar of the Dominican Province of the Philippines and the first Filipino rector of The Royal and Pontifical University of Santo Tomas.

He elected President of the Catholic Bishops' Conference of the Philippines in July 1986 and re-elected for a second term in July 1989.
