State Road 727 is a very short east-west Indiana state highway lying entirely within Steuben County, connecting Pokagon State Park to State Road 127 and hence Interstate 69. Termini are at the gate of Pokagon State Park and Indiana 127 at a rural intersection.

Until 1967 when Interstate 69 was completed in northernmost Indiana, it was designated as State Road 127, and State Road 727 referred to another short, now deleted highway in northeastern Indiana. Both incarnations of Indiana 727 are or were "child" routes of the old U.S. Route 27 which has been deleted in favor of Interstate 69.
