State Route 49 is a north-south state route located in the central part of the U.S. state of Georgia. The route travels from State Route 45 north of Dawson to State Route 22/State Route 24 and State Route 112 in Milledgeville.

From its southern terminus at State Route 45 north of Dawson, State Route 49 travels northeast to meet U.S. Route 280/State Route 27. State Route 49 becomes cosigned with these two routes, and they travel east to Americus, briefly becoming cosigned with U.S. Route 19/State Route 3, then State Route 30. In the eastern part of the city, State Route 49 departs U.S. Route 280 and continues northeast, through Andersonville, Oglethorpe, Montezuma, and Fort Valley. In Byron, State Route 49 has an interchange with Interstate 75 before continuing a short distance northeast to U.S. Route 41/State Route 11. The three routes become cosigned and travel north, and are later joined by U.S. Route 129/State Route 247. After entering Macon, U.S. Route 41 and State Route 247 depart, and U.S. Route 41 Business, U.S Route 129, State Route 11, and State Route 49 continue north into Macon, where they are joined by U.S. Route 80 and State Route 22. After crossing the Ocmulgee River and having an interchange with Interstate 16, State Route 49 departs the other routes and continues northeast to Milledgeville, where it, along with State Route 112, meet their northern terminus at State Routes 22 and 24 in downtown.

State Route 49 varies widely from a sparsely traveled route with an Average Annual Daily Traffic of less than 2,000 vehicles near its southern terminus with State Route 45 to over 20,000 vehicles or more around Macon. [1]In addition, State Route 49 between its intersection with State Route 49 Connector and Interstate 75 is part of the Fall Line Freeway, an expressway connecting Columbus and Augusta.

State Route 49 Connector is actually a bypass to the north and west of Fort Valley, connecting State Route 96 west of the city with the State Route 49 mainline northeast of the city. This route is also part of the Fall Line Freeway.
