State Road 61 is located in the Big Bend area of Florida, running through the state capital of Tallahassee.

The route originates at the Georgia border and is concurrent (and unsigned) with US 319 southwards until just north of I-10, where the routes separate and SR 61 continues south into downtown Tallahassee as Thomasville Road. It then briefly joins with US 27 as Monroe Street until that road branches off; 61 contines south to Paul Russell Road where it and State Road 363 'swap roads', SR 61 crossing over to Adams Street and continuing south as Crawfordville Road. At Capital Circle US 319 rejoins SR 61; just south of there SR 61 branches off as Wakulla Springs Road.
The first segment of State Road 61 terminates at the Wakulla County line, and the road continues south as Wakulla County Road 61. At the terminus of Wakulla Springs Road at Shadeville Highway (former State Road 365), this route turns west (still signed south) and runs to Crawfordville, where it again rejoins US 319 and becomes State Road 61 again, although this second segment of SR 61, which ends at US 98, is not signed.
