Chameli is the story of a prostitute,(Kareena Kapoor). Aman Kapoor (Rahul Bose) an investment banker, lost his wife, Neha (Rinke Khanna) in a car accident on a rainy night. Neha was pregnant with their child. The accident left him depressed and lonely. He drowned his grief with alcohol or smoking. Chameli (Kareena Kapoor) is a prostitute who was sold to a brothel by her uncle when she was young. She is now a tough, street smart girl.

On one rainy night, Aman and Chameli, two strangers, meet each other in South Mumbai. Both of them met on the rainy night and have a terrible past. Initially hesitant, but both of them start conversation soon. Aman, 32 is smart, suave consultant, who has lost his wife while giving birth to his baby; Chameli's uncle sold her at a young age to a brothel. Both of these incidents happened on a rainy night and so they are terrified of the rain.

The story slowly starts to progress, but has a twist lied ahead. While Chameli asks Aman to spend the night with her, he is still not interested.
