The two-hour pilot opens with an underground motorcycle chase. Blade is chasing down a Russian officer. A fight ensues, and it is revealed that the Russian officer is a vampire. Blade is seeking information, and after being beaten in the fight, the Russian officer reveals that the person Blade wants is in Detroit. Blade kills the vampire / Russian officer.

A slimy police officer, Detective Brian Boone, from Detroit picks up a young hooker. He takes her to an abandoned warehouse, takes her purse, and leaves her for the vampires to devour.

A scruffy, dark haired man named, Zack is seen in the back seat of a car with a glamourous peroxide blonde woman named, Chase. In the front seat are Marcus Van Sciver and a street tough named, Fritz. Zack is excited, because he thinks he is finally going to be turned into a vampire. When the car stops at the top of a parking garage, Marcus tells Zack that he has been asking too many questions about the House of Chthon, a vampire organization. Instead of turning him into a vampire, Marcus shoots him in the head, killing him.

Cut to the young and beautiful Sgt. Krista Starr, recently discharged from the army. Krista has recently returned from the war in Iraq and arrives home to a party thrown by her family. When she looks around for her brother, her family is unsure of where to find Zack. An argument ensues, and we learn that Krista's mother has given up on her brother as a bad seed. Krista defends him as a good man who deserves another chance. A knock on the door brings bad news from police that there had been a shooting, and Zack was killed.

Krista identifies Zack's body at the morgue and then meets Detective Boone--the same sleazy detective who brought the hooker to the nest of vampires earlier. The scene then cuts to Zack's funeral, which was sparsely attended. Afterwards, Krista goes back to her brother's apartment to try to get some information that might help her understand why he was killed. Upon arriving, she spots a man moving around the room, who hurriedly jumps out of the window when he realizes that he has been spotted. She follows him. The man turns out to be, Blade. Krista sets off to track down information about her brother's death by finding Blade.

Meanwhile, at an unnamed medical facility, vampires are being wrapped in plastic and used in some kind of vaccine testing program. Vampires are being subjected to garlic injections, which cause anaphylactic shock so severe in vampires that it's lethal. The vaccine doesn't work, and the vampire is ashed

After some further investigation, Krista learns the origins of Blade, realizes that there really are vampires and werewolves around her. Her digging attracts the attention of some vampires, who stage a traffic accident to lure her to an abandoned warehouse. Once inside, she is attacked by vampires who attempt to kill her. She's saved by Blade, who tells her to leave and to stop digging into Zack's death. She demands to know more and offers to help, and Blade eventually relents, allowing her to join him with his hunt. He tells her that Zack was murdered by Marcus Van Sciver, a wealthy socialite vampire who has been making a name for himself in Detroit by revitalizing slums. Blade and Shen show Krista a transponder that they want to plant on Marcus. So, Krista attempts to trap Marcus at a party, but her cover gets blown and she gets captured by the female vampire, Chase, who delivers Krista to Marcus.

Marcus brings Krista back to his hotel and injects her with his blood, turning her into a vampire. He then throws her off the hotel's roof, killing her, so she can turn. Krista wakes up in the city morgue as a newly born vampire, and makes her way to the roof of the morgue, where Marcus is waiting with Fritz. Marcus welcomes Krista to the "family". After Krista drinks some stored blood with Chase, Marcus presents her with Detective Boone, her first kill, whom she attacks and bleeds.

Blade tries to rescue Krista, whose activities he had been monitoring, but winds up in a fight with Chase and Krista. Blade grabs Krista and drags her to the hallway where he injects his serum to "get her thinking straight" again. In the meantime, the Aurora vaccine has just been given to Marcus' main henchman, Fritz--the street tough that was driving the car earlier--and now he appears to be a super-vampire who is immune to garlic, silver, and sunlight. A climactic fight between Blade and Fritz leaves Blade victorious, but confused as to why his silver-based über-weapon did not kill Fritz. Shen protests that something must be wrong, because weapons he designs "always work".

Blade contacts Krista and gives her some of the serum he uses to control "the thirst" and asks her to serve as an insider in order to help him bring down Chthon, he also reveals that her brother, Zack, was working for him when he was killed by Marcus. Krista takes the serum and walks away. When Shen asks if Blade thinks she will help them or the vampires, Blade comments that she "could go either way."

Detective Boone wakes up after Krista bleeds him and is a vampire. As Boone starts to laugh at the irony of being a vampire after Marcus tried to dispose of him, the camera pans out, and we see that he has been captured and placed in the vaccine testing program as well.
