Season 5 of MADtv ended with Pat Kilbane and Phil LaMarr leaving the cast to pursue other ventures outside of sketch comedy. Retaining most of the original cast (Nicole Sullivan, Debra Wilson), with newcomers from seasons 3 and 4 (Alex Borstein, Mo Collins, Michael McDonald, Will Sasso, and Aries Spears), MADtv would enter its first season of the 21st century with the usual changes: a new opening sequence and cast changes (Last year's featured player Nelson Ascencio and newcomer Christian Duguay would be repertory players, while newcomers Andrew Daly and Stephnie Weir would be featured for this season).
Despite his one-year tenure, Christian Duguay would install solid comic timing to the balanced cast with celebrity impersonations of President George W. Bush, *NSYNC member Joey Fatone, and Jay Leno. Duguay, like Frank Caliendo in the next season, didn't have many recurring characters to lean on, except for the hyperactive preteen Edward, and Loopgarue of "Loopgarue & Hulahoop," which he would team up with Mo Collins as the latter titular character.
Additionally, Jeff Richards would not be featured in MADtv for long, but he would be picked up by Saturday Night Live's executives and became well-known for his "Drunk Girl" character during his 3-year tenure on SNL (2001-04.)
Aside from these changes, MADtv continuously entertained the same way. Cast members Will Sasso and Alex Borstein continued to interview celebrities at the red carpet of big-time events, be it movie openings or award galas. Michael McDonald formed another infamous character, The Depressed Persian Tow Truck Man (later would be named Mofaz the following season), an irritable immigrant who whines about his family and uses pop culture references to further discuss how bad they are. Mo Collins kept appearing as Doreen Larkin, Lorraine Swanson, Trina Moss, and Liz-Whitman Goldfarb. Will Sasso still was called to perform more outrageous celebrity impersonations (this year, he did Fred Durst of Limp Bizkit and Robert De Niro). And season 5 guests Seth Green and Susan Sarandon popped up again to play characters old and new.

The MADtv logo appears. The theme song, which is performed by the hip-hop group Heavy D & the Boyz, begins. Each member of the repertory cast (followed shortly by the featured cast) are introduced, alphabetically, in the following manner. First, after MADtv logo appears, the screen is totally divided into three live action color clips of one cast member. The clips are depictions of the cast member in several of his/her recurring characters/celebrity impressions. Then the three clips divide into multi clips that take up the whole screen. Next the multi clips are replaced by one color still photo of that cast member. When the last featured cast member is introduced, the music stops and the title sequence ends with the phrase "You are now watching MADtv..."

- This was the last season to feature original cast member, Nicole Sullivan. Sullivan would, however, make several guest appearances on the show in later seasons.
- This was also the last season for Nelson Ascencio and the only season for Christian Duguay.
- Out of the four featured players of Season 6, only two members, Andrew Daly and Stephnie Weir were retained and promoted to repertory status for the following season. Dannah Feinglass's contract simply was not renewed. However, Jeff Richards left MADtv to be a featured player on Saturday Night Live, thus becoming the first performer to have been a cast member on MADtv and SNL.
