Stritch was born in Nashville, Tennessee, one of eight children to Garrett Stritch (the manager of Sycamore Mills in Nashville) and Catherine Malley, both of Irish ancestry. He was ordained a priest by Cardinal Pietro Respighi on May 21, 1910 in the Nashville. On August 10, 1921 he was appointed bishop of Toledo in Ohio, where he was consecrated a bishop by Archbishop Henry Moeller on November 30 later that year. He was appointed archbishop of Milwaukee on August 26, 1930 and installed November 19 before being appointed archbishop of Chicago on December 27, 1939 and installed January 3, 1940.

He received the red hat (with the title of S. Agnese fuori le mura) on February 18, 1946. On March 1, 1958, he was appointed Pro-Prefect of the Congregation for the Propagation of Faith, thus became first American member of the Roman Curia. He was called to Rome, where he died at the age of 70 on May 27, 1958.

He is interred in the Bishops’ Mausoleum at Mt. Carmel Cemetery, Hillside, Illinois. Cardinal Stritch was succeeded as Archbishop of Chicago by Albert Cardinal Meyer; and as Pro-Prefect of the Congregation for the Propagation of the Faith by Grégoire-Pierre Cardinal Agagianian. Entertainer Elaine Stritch is his niece. Cardinal Stritch University in Milwaukee and Cardinal Stritch High School in Oregon, Ohio are both named for him.
