Pennsylvania Route 16 is a 43.1 mile long east-west state route located in southern Pennsylvania, United States. The western terminus of the route is at U.S. Route 522 in McConnellsburg. The eastern terminus is at the Mason-Dixon Line in Liberty Township, where PA 16 continues into Maryland as Maryland Route 140. The road's main name is Buchannan Trail.

PA 16 begins at an intersection with U.S. Route 522 in McConnellsburg. PA 16 begins as Lincoln Way and makes a 45 degree turn towards the southeast as Buchannan Trail leaving the boro of McConnellsburg. In Franklin County, PA 16 passes by Buchannan's Birthplace State Park where former president James Buchannan was born. In Peters Township, PA 16 intersects the northern terminus of Pennsylvania Route 456. PA 16 continues towards the east and southeast. In Mercersburg, PA 16 is known as Main Street and begins a three route concurrency with Pennsylvania Route 75 and Pennsylvania Route 416. At the Mercersburg boro line, PA 75 leaves the concurrency and PA 16/PA 416 continue concurrent as Buchannan Trail towards the southeast. In Montgomery Township, PA 416 splits from PA 16 and PA 16 continues towards the east. At the eastern tip of Montgomery Township, PA 16 begins a concurrency with Pennsylvania Route 995. In Peters Township, PA 995 splits from PA 16 in the hamlet of Upton. In Greencastle, PA 16 is called Baltimore Street and intersects U.S. Route 11. Easto of US 11, PA 16 intersects Interstate 81 at exit 5. PA 16 continues towards the east as Buchannan Trail. In Waynesboro, PA 16 has two pair of concurrencies and the first one is is with Pennsylvania Route 316. The second concurrency is with Pennsylvania Route 997. PA 16 is called Main Street in Waynesboro. In Carroll Valley, PA 16 is renamed to Waynesboro Pike and PA 16 intersects the western terminus of Pennsylvania Route 116. PA 16 terminates (ends) at the Maryland border (Mason-Dixon Line) and it continues as Maryland Route 140.

PA 16 was signed in 1928.

From 1925 to 1967, present-day PA 16 from US 522 to the Buchanan Trail/Lincoln Way intersection in McConnellsburg was occupied by Pennsylvania Route 1 (from 1925 to 1930) and its successor, U.S. Route 30 (from 1926 to 1967). In 1967, US 30 was rerouted along a bypass north of town, allowing PA 16 to be extended westward to its present terminus at US 522.[1]
