New York State Route 305 is a north-south route in the Southern Tier that runs from the Pennsylvania state line in Cattaraugus County to the hamlet of Belfast in Allegany County, where it ends at NY 19. It intersects the NY 17/Interstate 86 Southern Tier Expressway at Cuba.

305 was once the designation for the current course of NY 45 in Rockland County, which explains why it's the only route number, commissioned or decommissioned, between 299 and 308 used somewhere in New York outside the Hudson Valley.

The section south of Portville was once part of NY 16. It was added to 305 when 16's entire course south of Olean was relocated westward.
