New York State Route 261 is a north-south running highway located entirely in Monroe County to the west of the city of Rochester. The southern terminus of the route is located at NY 104 in Greece. Its northern terminus is located at the Lake Ontario State Parkway in Manitou Beach.

At NY 104, Manitou Road, Monroe County Route 208 and the highway that NY 261 follows for most of its length, loses the County Route 208 designation and becomes NY 261. Proceeding north from 104, NY 261 maintains a near-perfect north-south alignment until it enters the Manitou Beach area. Along the way, NY 261 joins with NY 18 for a short, 0.4 mile duplex in West Greece.
Near Manitou Beach, Manitou Road branches into two roads, both of which eventually intersect with the Lake Ontario State Parkway. The left branch, which continues on the same north-south alignment as Manitou Road, becomes Payne Beach Road while the right branch, which turns to the northeast, becomes Manitou Beach Road and carries NY 261. Shortly afterward, NY 261 arrives at its northern terminus at the L.O.S.P.
An interesting note is that NY 261 is the westernmost exit on the limited-access portion of the Parkway in Monroe County. After interchanging with Manitou Beach Road, the Parkway becomes a four-lane divided highway until reaching Orleans County, where it reverts to a limited-access highway.
