Route 72 is a state highway in the U.S. state of New Jersey. It runs 28.74 miles (46.25 kilometers), heading westward from Ship Bottom on Long Beach Island in Ocean County to an intersection with Route 70 in Woodland Township in Burlington County.
Approaching Long Beach Island, Route 72 crosses Manahawkin Bay via the Dorland J. Henderson Memorial Bridge.
Plans were in place for a proposed Route 72 freeway from the western terminus at Route 70 to the concurrently proposed Interstate 895, connecting Philadelphia's northern suburbs with the Jersey Shore. The freeway plans along with I-895 were canceled in 1981.

Prior to the 1953 renumbering, Route 72 was Route S40.
In 1968, a new alignment of Route 72 was built bypassing Bay Avenue east of Manahawkin. Bay Avenue was then renumbered Route 180. It was decommissioned in 1980, and became County Route 680. It was later decommissioned by Ocean County as well, and is now locally maintained.
