Former Route 52 was one of the original 1922 highways. Its northern terminus was at Route 1 in St. Joseph, its western terminus was at the Kansas state line east of Atchison, Kansas. In 1926, it became part of U.S. Route 59. A branch line, Missouri 52A, ran from Missouri 52 to the Buchanan/Platte County line. That road is now Route 273.
