Mark Kerr (or Ker) (1553 - 1609), 1st Earl of Lothian was a Scottish nobleman and politician.

The son of Mark Kerr, abbot of Newbattle, Midlothian, he was appointed Master of Requests in 1577 made a baron and Privy Counsellor in 1587, and created a Lord of Parliament in 1591. He acted as interim Lord Chancellor of Scotland in 1604. He was created Earl of Lothian in 1606. He resigned the office of Master of Requests in 1606.
