The year is 2003 and the class of 1953 is having their 50th reunion at the Pandora Hotel. Tyler Perry is known for his improvisation, so audiences may see different versions of the production. However the general synopisis is as follows

The stage play is divided into two Acts.

Act I opens up with Cora as an employee of the Pandora Hotel. Her employeer, Ann, learns that Cora's mother, Madea will be staying at the hotel and offers to upgrade her room. Perry then enters as Dr. Willie Leroy Jones who is hired as the bellman and the bartender, who is trying to earn money for his rent. Miss Emma is a 74 year old housekeeper, who is ready for retirement. She's worked at the hotel for 34 years. But her daughter, Stephanie has left her two children on her doorstep, leaving Miss Emma having to work in order to support them. Stephanie is on drugs. Also staying at the hotel is Brown, who carries the remains of his recently deceased wife in a Pringles can. When Brown tries to shout, Ann replies, "We do not do that here." Mr. Clarence Massey also checks into the hotel, who is Diana's husband.

From here, the story line begins to unfold. Stephanie comes into the hotel paranoid, needing a room, but doesn't have all the money. Cora offers to help her out, but only for 1 night. Then Stephanie runs into her mother, Miss Emma. Miss Emma asks her when she is going to get the kids, but Stephanie gives her the runaround because she is hiding from her pimp husband, Horace.

As Clarence and Ann are talking at the bar, Ann tells him that his son misses him. But Clarence says that even though he promised to leave his wife that weekend, but his wife has cancer and she would be coming into the hotel in a wheel chair. Willie starts to make jokes. They leave together and go upstairs.

Next Brown and Stephanie come to the bar. Stephanie invites Brown up to her room. Willie warns Brown that she's a hooker and to have his wallet ready. But naive Brown thinks she is a wholesome girl.

Corey Jeffrey and his wife Trina are the next couple to check into the hotel. Willie stirs up trouble between the couple when he calls Trina's name and pretends that he knows her. Corey and Trina are on a get-a-way to try and repair their marriage after Trina was unfaithful. Corey, however, refuses to forgive her.

Diana, who is celebrating her 10th wedding anniversary, checks into the hotel. She is an old friend of Cora's who hasn't been to church in 10 years. Willie makes the connection that Clarence lied about her having cancer. Willie rushes Diana upstairs so they can catch Ann and Clarence in the act, but are seconds late. Willie makes it clear to Clarence that he knows his secret. Clarence gives Willie money, his jogging suit and pants, his Jordans and watch in order to keep his mouth shut.

Brown comes back downstairs with Stephanie, after he had been praying for her the entire time. Then Horace enters the hotel and sneaks up on them. When Horace tries to make Stephanie leave with him, Brown tries to step in but Horace scares him off. Stephanie refuses to leave. Horace tells Stephanie to wave goodbye to their kids outside. Stephanie recultantly leaves with Horace.

After Willie's his many breaks, his humorous remarks, and his blackmailing of Clarence, Ann fires him. Willie then quits and agrees to leave. But not before Ann is introduced to many the 27 different personalities that live in Willie's head after she refuses to give him his check.

Ann isn't through yet. She fires Miss Emma to make herself look good. Her excuse is that she's running a business that could care less about what Miss Emma thinks she deserves after her 34 years of employment. Now Miss Emma is left with no job, two grandbabies to care for, and a hooker daughter to pray for.

Spoilers end here.
