M-49 is a short north-south route in south-central Michigan that goes from Litchfield south to the Ohio state line. It passes through Allen where it crosses US 12 (the old US 112). It also passes through Reading, and Camden. Near the Michigan/Ohio state line just south of Camden, it has a junction with Territorial Road, a Hillsdale county road that becomes SR 120 as it crosses the Indiana state line in the short segment of north-south border between Indiana and Michigan.

Note: M-49 is the only Michigan highway in which the road crossing into another state has the same state highway number. M-49 continues as SR 49, which is the westernmost interchange with the Ohio Turnpike.
