- Shin'ichi Kudo (工藤 新一 Kudō Shin'ichi?) / Jimmy Kudo (Voiced by: Kappei Yamaguchi (Japanese), Jerry Jewell (English))- A high school detective now living in the body of a small boy under the alias Conan Edogawa (江戸川コナン Edogawa Konan?) (Voiced by: Minami Takayama (Japanese), Alison Viktorin (English)).

- Skilled at: Football (soccer) and mystery novels,acting
- Ran Mori (毛利 蘭 Mōri Ran?) / Rachel Moore (Voiced by: Wakana Yamazaki (Japanese), Colleen Clinkenbeard (English))- Jimmy's best friend since elemetary school. Both are in love with one another but neither had the courage to admit their feelings. She is skilled in the martial arts and is the captain of her karate team, though her resolve can be easily broken if she has to face anything related with the horror genre. She spends most of her home life looking after Conan (whom she does not know is really Jimmy) and her irresponsible father, Detective Richard Moore. She sometimes threatens him with a punch on the table, but is a very genial person.

- Kogoro Mori (毛利 小五郎 Mōri Kogorō?) / Richard Moore (Voiced by: Akira Kamiya (Japanese), R. Bruce Elliot (English))- Rachel's father, an incompetent private detective. He's married to Eva Kadan (Eri Kisaki in the original Japanese anime and manga), a successful lawyer. They are now living separately. Despite his irresponsible ways, he appears to care deeply for his daughter, and at times shows some flashes of honor and strict ethics.

- Hiroshi Agasa (阿笠 博士 Agasa Hiroshi?), also Agasa-hakase, which means Doctor Agasa) / Dr. Herschel Agasa (Voiced by: Kenichi Ogata (Japanese), Bill Flynn (English))- An absent-minded professor who invents several devices to help out Jimmy. His given name is Hiroshi in the English manga and Herschel in the English anime. He serves as one of the few roles in the story who learns about Conan's true identity and helps the little detective in cases.
- Skilled at: inventing weird machines, but is not always successful
- Shiho Miyano (宮野志保 Miyano Shiho?), also known as "Sherry" (シェリー) and as Ai Haibara (灰原哀 Haibara Ai?) (Voiced by: Megumi Hayashibara)- A former member of the crime syndicate that created APTX 4869, the poison that shrank Jimmy. Shiho betrays the organization after Gin killed her sister, Akemi Miyano. Shiho took the drug in a suicide attempt, but rather than dying, she, like Jimmy, became small. She is taken in by Dr. Agasa and becomes Conan Edogawa's right hand under her alias Ai Haibara. She shows affection toward Jimmy and is cheered by him as well, but she denies having said feelings.
- Skilled at: Biological Research,acting (through out most of the episodes and movies, she will do anything for Jimmy Kudo/Edogawa Conan's sake no matter how tired she is. Also, she is able to make herself cry, a trick Jimmy is incapable of, if only because he's a man).

- Ayumi Yoshida (吉田歩美 Yoshida Ayumi?) / Amy Yoshida (manga) or Amy Yeager (anime) (Voiced by: Yukiko Iwai (Japanese), Monica Rial (English))- A girl who becomes friends with Conan, Amy is the token female member of the Junior Detective League. She has a crush on Conan (which makes him uncomfortable, because he, of course, loves Rachel, though Ayumi thinks that he loves Ai Haibara because the first person who he is protect is Ai). A naive and innocent little girl who shows courage from time to time and acts as the team's cheerful spirit.
- Mitsuhiko Tsuburaya (円谷光彦 Tsuburaya Mitsuhiko?) / Mitch Tsuburaya (manga) or Mitch Tennison (anime) (Voiced by: Ikue Ootani (Japanese), Cynthia Cranz (English))- A friend of Amy who has a scientific approach. He seems to be infatuated with both Amy and Ai Haibara (he doesn't know who he should protect). A smart and well-read first-grader who believes that technology can solve all problems.
- Genta Kojima (小嶋元太 Kojima Genta?) / George Kojima (manga) or George Kaminski (anime) (Voiced by: Wataru Takagi (Japanese), Dameon Clarke (English))- The overweight, self-proclaimed leader of the Junior Detective League (Detective Boys, or Shōnen Tantei, in the original Japanese anime and manga). He has a crush on Amy.

- Juzo Megure (目暮十三 Megure Jūzō?) also known as Inspector Megure (目暮警部 Megure-keibu) / Joseph Meguire (Voiced by: Chafurin (Japanese), Mark Stoddard (English))- A veteran police officer who has been working with Conan, Yusaku Kudo and Richard Moore; in fact, Moore used to work under him 10 years ago. It was noted in the second 'Detective Conan' movie that he never took his hat off; it is eventually revealed in Volume 29 of the manga that he uses it to hide an old scar which reminds him of a special case from his early police days.
- Miwako Sato (佐藤美和子 Satō Miwako?) (Voiced by: Atsuko Yuya)- A young female officer who works with Meguire. Her father was also a police officer who was killed on duty when she was only six years old. Her former partner Matsuda was killed in an explosion. She made her debut as a cameo appearance in Volume 19 of the manga and was properly introduced in Volume 21. Tough, energetic, and a dedicated police officer (so dedicated that her "dates" tend to turn into police investigations) she is hugely popular with the male members of the police department, but - to the latters' chagrin - she, albeit hesitantly, begins to develop feelings for her colleague Wilder.
- Wataru Takagi (高木ワタル Takagi Wataru?) / Detective Wilder (Voiced by: Wataru Takagi (Japanese), Doug Burks (English)- Another officer that works with Meguire and has a crush on Satou which is - if somewhat arkwardly - requited. He was first introduced in the anime and made his first manga appearance in Volume 17. He has an appearance of ineffectual nervousness, but he's not as useless as he seems. His quick ability to assess a situation and take immediate and decisive action when necessary are points in his favor. He also appeared to know (or at least suspect) about Conan/Jimmy's secret identity and problem; this is seen when he makes close observation to Conan in some investigation and in the end of Volume 36 (when Conan wants to defuse the time bomb).
- Officer Chiba (千葉刑事) - Wilder's partner who likes to eat a lot. He's nice and very encouraging towards his friend. He made his debut in the fourth Detective Conan movie, then as a cameo appearance in Volume 29 of the manga and was properly introduced in Volume 32. Later in the series it's revealed he is a fan of the children's show Kamen Yaiba (Case Closed's counterpart to Japan's "Kamen Rider").
- Ninzaburo Shiratori (白鳥任三郎 Shiratori Ninzaburō?) (Voiced by: Kazuhiko Inoue)- Another officer who works with Meguire and likes Satou; however, she does not feel the same about him. He made his first appearance in the first Detective Conan movie and his manga appearance was in Volume 21, along with Officer Satou. Later in the series he makes overt advances towards Miwako, relying heavily on his status as the scion of a wealthy family (and doing his best to make the shy and awkward Wilder feel inferior in the process).
- Police Superintendent Kiyonaga Matsumoto is the superior officer of Juzo Megure and his underlings. His brutish appearance and the scar running down his left eye easily hide the fact that he is a caring, if somewhat stern, father figure. He is widowed and has a single daughter, Sayuri, who used to be Shinichi's, Ran's and Sonoko's music teacher in middle school.
- Inspector Sango Yokomizo (横溝 参悟 Yokomizo Sango?) / Detective Worthington - a stern-looking police inspector from Saitama whom Conan has run into on several occasions. He is a serious and capable police officer, though he tends to be sceptical about theories and to intimidate suspects by moving up close and staring at them. His twin brother Jugo (also an inspector) works at the Kawanaga police department. He made his first appearance in the Case Closed volume #6.
- Inspector Yamamura - a young police officer from the Gunma Prefecture. Thanks from watching too much TV, he is very enthusiastic about his work but totally naive, bumbling and child-headed, and thus totally unsuited for the job. The first reaction of Conan and all who know him is exasperation whenever they happen to encounter him; but once set on the right track he is very supportive.
- Jinpei Matsuda (松田陣平 Matsuda Jinpei?) - Miwako's parter who died while trying to stop a bomb from exploding. He chose to stay inside a Ferris wheel ride with a bomb because a message came that another bomb's location would be displayed on the casing 3 seconds before the explosion. He originally meant to redeem his partner Hagiwara, who died 4 years before in a related incident, but was unable to succeed. Miwako was partnered with him for less than a week, but his intense personality and subsequent death left a huge and lasting impact on her for years to come.
- Yumi Miyamoto (宮本由美 Miyamoto Yumi?) - Traffic officer who knows about Wilder's crush on Sato and often teases the two about their relationship. She also has two dogs.
- Inspector Otaki - a member of the Osaka police department who is also a close friend to Harley Hartwell and has learned to trust him in the face of hard-to-crack cases.
- Heizo Hattori (服部 平蔵 Hattori Heizō?) - Harley's father is the chief commissioner of the Osaka police department. While a very serious and stern police officer and family man, he has come to admire his son's abilities, though he rarely shows.
- Commissioner Toyama - Kazuha Toyama's father and member of the Osaka police department who works closely with Heizo Hattori.

- Further information: Black Organization (Case Closed)
- Gin (Voiced by: Yukitoshi Hori) - One of the top-ranked members in the BO, and the person who gave the APTX 4869 poison to Jimmy. Extremely cold-blooded and one of the most menacingly-drawn of the villains. It should be noted that he is also extremely perceptive, which explains why it would be extremely difficult to catch him.
- Vodka (Voiced by: Fumihiko Tachiki) - A member of the BO; always seen alongside Gin and refers to him as "Aniki" or older brother.
- Tequila - A giant BO henchman towering above 6'5". Introduced in volume 12 of the manga (episode 54 of the anime), and accidentally killed by a briefcase bomb in the same volume/episode.
- Vermouth (Voiced by: Mami Koyama) - An important member of the Black Organization. Her true identity is Sharon Vineyard, a famous American actress who learned the skill of disguise alongside Vivian Kudo, Jimmy's mother. She is the Boss's favorite member of the BO.
- Kir (Voiced by: Kotono Mitsuishi) - Originally a newscaster named Rena Mizunashi, Kir also serves as a member of the Black Organization, as revealed in episode 425 (volume 48-49 of the manga). She is knocked unconscious and detained by FBI agents after being involved in an accident. It is revealed that her name is most likely an alias that refers to 007.
- Chianti - A member of the Black Organization who serves as an assassin and replaces Calvados. She appears in volume 48-49 of the manga and episode 425 alongside her partner, Korn. She has a tattoo on one of her eyes. Chianti does not appear to like Vermouth.
- Korn - Chianti's partner. Appears in episode 425 and volume 48-49 of the manga. He is much older than his comrades and wears dark sunglasses. Unlike the other members of the Black Organization, he never smiles.
- Calvados- Sniper who aids Vermouth in almost killing Jodie, but near the end of episode of 345 he eventually killed himself rather than being captured.
- Kusuda Rikumichi- A member of the Black Organization who posed as a patient in hopes of tracking Rena Mizunashi; however, Conan and the FBI managed to find him out. He escaped the hospital, and Conan and Shuichi went after him, but he committed suicide for unknown reasons before they managed to get information. He first appears in volume 57 of the manga.
- Boss - Leader of the Black Organization. Unseen and unheard, known only through text messages. His phone number follows the tune of the children's song "NANAZU NO KO (7 year-old child)".

- Jodie Starling - First appeared in episode 176 as Rachel's English teacher under the name of Jodie Saintemillion. She appeared to know Conan's secret, judging by her dialogue towards him (calling him "cool kid," etc). For a long time she was suspected by Conan friends to be Vermouth in disguise, and even investigated by Conan and Harley on these suspicions in episodes 277-278, but is later revealed to be an FBI Agent in episode 345. She has a personal vendetta against Vermouth concerning the death of her parents. Please note that her name is a reference to Agent Clarice Starling from "Silence of the Lambs" played by actress Jodie Foster.
- Shuichi Akai (赤井秀一 Akai Shūichi?) (Voiced by: Shuuichi Ikeda)- First appeared in episode 230-231 as a mysterious man spying on Conan. He is later revealed to be an FBI agent in episode 345 and Jodie's partner. In the manga, Gin refers to him as a very dangerous man after Vermouth informs him about her encounter with Conan and the FBI agents.
- James Black - Another FBI Agent who works with Shuichi and Jodie.
- Andre Camel- A new FBI introduced in Chapter 600. He works as Jodie's partner, but both Jodie and Conan are very suspicious of him. As of File 602, he knocks Jodie out by punching her in the stomach. His true loyalty and intentions are yet to be revealed.

- Sonoko Suzuki (鈴木園子 Suzuki Sonoko?) / Serena Sebastian (Voiced by: Naoko Matsui (Japanese), Laura Bailey (English))- Rachel's best friend. Sonoko is very obsessed with guys, but later she develops a somewhat genuine liking towards Makoto Kyougoku, a promising martial artist. Sonoko has a sister (named Ayako Sebastian in the English manga, Angie Sebastian in the English anime, and Ayako Suzuki in the Japanese versions). She thinks Jimmy is a loser and is always trying to set Rachel up on dates with new guys.
- Heiji Hattori (服部平次 Hattori Heiji?) / Harley Hartwell (Voiced by: Ryo Horikawa (Japanese), Kevin M. Connolly (English))- Jimmy's rival detective from Osaka, who later becomes friends with him. He has a relationship with martial artist Kazuha Toyama (遠山和葉 Toyama Kazuha), his childhood friend and would-be love interest, and this is pretty similar in the case of Jimmy and Rachel. He is also one of the few people who know that Conan is really Jimmy. Harley is also a master Kendo fighter. Unlike Jimmy, his favorite author is Ellery Queen rather than Sir Arthur Conan Doyle who wrote Sherlock Holmes.
- Kazuha Toyama (遠山和葉 Toyama Kazuha?) (Voiced by: Yuko Miyamura ) - Harley's would-be girlfriend and (like him) the child of a police officer. Her relationship with Harley closely mirrors Rachel's with Jimmy, because both are too shy to admit that they feel more than 'friendship' for their respective partner; perhaps this is what makes her a close friend to Rachel. She is skilled in Aikido.
- Yusaku Kudo (工藤優作 Kudō Yusaku?) / Booker Kudo (Voiced by: Hideyuki Tanaka (Japanese), Randy Tailman (English))- Jimmy's father, a famous author of detective stories. Created the popular character, "Night Baron".
- Yukiko Kudo (工藤有希子 Kudō Yukiko?) / Vivian Kudo (Voiced by: Sumi Shimamoto (Japanese), Laurle Steele (English))- Jimmy's mother, a former actress who gave up her career at age 20 to marry Yusaku against the will of her family. A Yukiko Kudo also appears in episode 4 of CLAMP School Detectives, another mystery anime. She has been a rival of Rachael's mother in her high school years, and was elected Miss Teitan High School. Because of her reputation as the wife of a famous detective author and for involving herself in some criminal cases, she has gotten the nickname "Night Baroness".
- Tomoaki Araide (新出 智明 Araide Tomoaki?) is a young doctor and the son of Richard's physician. While he had practised along with his father, he has moved to Aomori for a job at a local clinic, along with his grandmother Mitsu Araide and his household helper Hikaru Yatsumoto, especially after his father had been murdered by his stepmother Yoko for driving his actual mother (and Yoko's friend), Chiaki, into alcoholism and eventual death. However, he has reappeared in Conan's vicinity on several occasions - seemingly at least; his identity was used by Vermouth in order to track down Ai aka Sherry. He was once the basketball coach on Rachel's school and, upon their first encounter, thought to be a rival for Jimmy/Conan's affections for Rachel.
- Eisuke Hondou (本堂 瑛祐 Hondō Eisuke?)(Voiced by: Junko Noda )- is an odd and clumsy guy. He's Rachel's new classmate and shockingly looks more like a female rather than a male. He seems to have an awkward appearance, but deep down, he is undercover. In episode 462 (The Shadow of the Black Organization: The Young Witness), he accidentally takes a picture of suspects on Rachel's cellular phone in a tattoo shop, but Conan knows that he did it on purpose. Conan suspects he works for the Black Organization because of a technique he does similar to Kir, a woman working for the Black Organization. Conan is the only one having second thoughts about Eisuke, but others think that he's a just a smart misguided student. In episode 464 (The Shadow of the Black Organization: Strange Illumination) Eisuke tries to dig for information on how Richard Moore figures out his cases, but Conan cunningly saves himself by saying he is tired; therefore, Richard says he'll talk about his solved cases later. Swiftly, Eisuke glares at Conan; at that point, Conan realizes that his identity is at sake.
- Yoko Okino (沖野 ヨコー Okino Yokō?): a young pop star idol who is a regular focus of worship by Richard Moore. She has a number of cameos throughout the series, mostly as a prominent guest at some social gatherings which turn into criminal cases.
- Hikaru Yatsumoto is the young household keeper to Dr. Tomoaki Araide. She tends to be overly nervous about her chores, mostly because of the possibility that she may have a crush on her young employer (she used to be his patient once) and therefore is too anxious about having to leave him.
- Kaitou Kid (怪盗キッド Kaitō Kiddo?) (Voiced by: Kappei Yamaguchi (Japanese), Jerry Jewell (English)) / Phantom Thief Kid - A thief that looks a lot like Jimmy (and also has a girlfriend named Aoko Nakamori, who, coincidentally, looks a lot like Rachel). Despite the fact that Jimmy/Conan does not care much about investigating thefts, Kid can be considered his archnemesis, as he is the one criminal (beside some members of the BO) he can't seem to catch. Curiously, Kaitou discovers Conan's true identity, but keeps his secret, and sometimes Jimmy can even count on Kid's help to solve some of his cases (when the criminal is not Kid, of course). His codename in the police department is 'Thief 1412'. Kaitou Kid appears in his own manga series, Magic Kaito. Kid steals antiques and arts of high values in search of his father's murderer (who was Kaitou Kid before he died 8 years ago).
