State Route 372 begins with its intersection with S.R. 140 in the western part of Alpharetta, Georgia and travels to the north, through unincorporated, but rapidly growing, areas of Fulton and Cherokee counties.
After intersecting with S.R. 20 and S.R. 369 in Free Home, the route turns more towards the northwest, crossing the Etowah River, then entering Ball Ground. In Ball Ground, the route begins a multiplex with S.R. 5 Business. In Nelson, the two routes utilize a short expressway stub before the routes end at S.R. 5 and S.R. 515.
