East Shore Hospital opened in the 1930s as the Paglar Maternity and Nursing Home, and was founded by Dr Palgar, a Eurasian general practitioner. In 1970, his descendants sold the hospital to an Englishman, Dr Leo Taylor and was renamed Saint Mark's Maternity Hospital.

Two years later, the hospital was sold to Hong Leong for residential purposes. However, the land was zoned for hospital used. Another doctor, Dr. Sundarason and a group of businessman and doctors in 1974.

The new management modernised the hospital's facilities and then commenced operations later that year as a 45 bed Saint Mark's Hospital. Three years later, Dr Yahya Cohen together with American Medical International entered a joint venture with the former to form Saint Mark's AMI Private Limited in May 1979.

Saint Mark's further purchased a 40,000 ft2 site adjacent to the existing building in 1982. It also increased to 49 beds as well. In 1983, AMI took over the facility and was renamed The American Hospital of Singapore which it then began to offer some of its speciality services. A medical office building to house speciality clinics was completed in March 1987 and the tenants moved into the building.

The National Medical Enterprises acquired the hospital in 1989 and was renamed to its current name. Parkway Holdings acquired both East Shore and Mount Elizabeth Hospitals in 1995 making Parkway the largest private healthcare organisation in the country.
