Leonardo: Things are not good, and they're getting worse by the minute. We could really use good old Donnie right about now. Ever since Bishop's mutant outbreak began, Don's been the one guiding us through this whole mess. There were plenty of times where we would've been turtle-waxed if Don's brains hadn't bailed us out. It's too bad Don's not here now, because this is definitely one of those times.

The episode opens with April answering the door and letting Casey in. Casey is all dressed up and ready to go to a classy French restaurant, while April was all geared up for a romantic bike ride. There is another knock on the door - it's the Turtles, who are going on another bug hunt, but want to leave Donatello behind, who is seriously ill. April agrees, much to Casey's annoyance and they look after Donatello. Meanwhile, Agent Bishop has retrieved Baxter Stockman's remains and has managed to restore him as a cyborg version of himself. While fighting bugs at a power plant, Michelangelo destroys one of the generators, cutting off the electricity for the entire city. Donatello's illness is starting to get worse as the lights go out. When April and Casey go off to find torches, Donatello begins to undergo a secondary mutation and manages to run away. When Casey and April find him in the basement, he has already fully transformed, and attacks Casey and April. April manages to get in touch with the other turtles, who arrive and chase Donatello out. To their shock, they realize that the monster was Donatello, and they decide to look for him before he hurts himself. They go to Leatherhead's lair in the old railway station, where Leatherhead discovers that the sample from Donatello's bandage is similar to the DNA of the mutants caused by Agent Bishop's outbreak. With their bug hunting gear, they decided to go and capture Donatello, using Michelangelo as bait. They successfully capture him and bring him back to Leatherhead's lair, where the Turtles, Splinter and Leatherhead vow to find a cure for Donatello.

- This episode was broadcast as the 100th episode; however, Tale of Master Yoshi is considered the 100th episode in the series chronology.
- This episode references and uses footage from the earlier episode Outbreak, in particular the moment in which Donatello is stung by a mutated bug.
- A brief scene in this episode reveals that Agent Bishop has recovered Baxter Stockman's brain from the bottom of the river (where it ended up in the episode Insane in the Membrane) and created a new, frail cyborg body to hold it.
- Donatello's appearance, when he undergoes his secondary mutation, is similar to that of Raphael's in Vol.4 of the Mirage Comics, when Raphael is bitten by vampires.
