The Jaguars opened the regular season against the Dallas Cowboys on September 10. After a slow start for the Jaguars, they pulled off a victory, in the process of spoiling the NFL return of WR Terrell Owens. The Jags performance was highlighted by the defense, three different players intercepting Dallas quarterback Drew Bledsoe. Byron Leftwich threw one touchdown and ran for another, while veteran running back Fred Taylor scored on a short run to effectively seal the victory early in the fourth quarter. Jags DE Reggie Hayward suffered what is considered to be a season ending Achilles injury.

The Jaguars played their second game at home against the defending Super Bowl champion Pittsburgh Steelers on Monday Night Football. The Jaguars won in what was the lowest scoring Monday Night game ever, 9 to 0. The Jaguar defense held the Steelers' running game to only 26 yards and allowed them to cross midfield only once. Steelers quarterback Ben Roethlisberger's passer rating at the end of the game was a measley 38.7. The defending Super Bowl champions have not been shut out since 1985. In their last meeting, the Jaguars defeated the Steelers 23-17 on October 16, 2005. The win improved the Jaguars record to 2-0.

The Jaguars played their first road game against the Indianapolis Colts on September 24. The game started out with the Jaguars gaining 105 rushing yards in the first quarter as well as a rushing touchdown by QB Byron Leftwich. In the second quarter the Colts' Terrence Wilkins scored on an 82 yard punt return. Two missed field goal attempts by Jaguars kicker Josh Scobee hurt the team. The Jaguars scored late in the fourth quarter and stopped the Colts from running the clock down. With about a minute left to play the Jaguars tried to score but were halted after Mike Doss intercepted Leftwich's pass. The Colts took a knee and handed Jacksonville their first loss of the season.

The Jaguars played the Washington Redskins on October 1 and were defeated in overtime by a touchdown scored by Santana Moss catching a 68 yards pass from Mark Brunell, the former Jaguars' quarterback. The Jaguars had a chance to win during regulation after a fumble recovery by free safety Deon Grant. But the celebration that ensued in the end zone with Marcus Stroud cost the team a 15 yards penalty, and they had to back out of the end zone only to settle for a field goal to tie the game. At the end of the game he recognized that he cost his team the chance to win the game during regulation.
