West Virginia Route 527 is a north-south route extending from WV 152 just south of Huntington, West Virginia to State Route 7 in Chesapeake, Ohio by way of the Sixth Street Bridge (maintained by the West Virginia Department of Transportation, the bridge's northern approach is designated State Route 527 by the Ohio Department of Transportation). While the current Sixth Street Bridge was opened to traffic in 1995, its predecessor once carried US 52 across the Ohio River to State Route 7; hence the current numbering.
The southern terminus of WV 527 is most unusual: it also serves as the northern terminus of WV 152, which continues along the same street as it passes over Interstate 64 (both state routes are indicated on exit signage on the expressway). WV 152 continues along the former US 52 path until its intersection with US 52 four miles north of Crum.
