U.S. Highway 150 is a 571 mile (919 km) long northwest-southeast United States highway, signed as east-west, running from Mount Vernon, Kentucky to Moline, Illinois.

- The highway's eastern terminus is in downtown Mount Vernon at an intersection with U.S. Highway 25. Proceeding northwest through Kentucky, Route 150 crosses the Martha Layne Collins Blue Grass Parkway at Bardstown and is multiplexed with U.S. Highway 31E north of there into Louisville. In Louisville, Route 150 is called Bardstown Road, intersecting Interstate 264 and Broadway and intersects Interstate 65. Route 150 is multiplexed with Interstate 64 briefly to cross the Ohio River on the Sherman Minton Bridge.
- Route 150 remains multiplexed with I-64 into Indiana and splits off west of New Albany, before proceeding roughly west from there. The route is multiplexed with U.S. Highway 50 from Shoals to Vincennes, before turning north and becoming multiplexed with U.S. Highway 41 through Terre Haute.
- The highway enters Illinois near Vermilion and proceeds northwest through Danville, Urbana and Champaign, Bloomington, Peoria, Galesburg and Moline. Between Danville and Moline, Route 150 closely parallels Interstate 74. The highway's western terminus is near the Quad City International Airport at an intersection with U.S. Highway 6 in Moline.
